How Stefan Mandel Won the Lottery 14 Times

The chances of winning the lottery are one in a million. But not for Stefan Mandel. In the 1980s and 1990s, the Romanian-Australian economist managed to win the lottery an astounding 14 times. Everything he did was perfectly legal and all it took was some simple calculations.

Mandel was born in Romania and his first two wins came in his home country. He was spurred to win the lottery to collect enough money to move his family out of the then-Communist country. To do so, he developed an ingenious system. First, he looked at different lotteries to see how many different combinations were needed to cover every possible draw.

For instance, if a particular lottery plays numbers 1 to 40 and requires a six-number combination to win, there are 3,838,380 possible outcomes. Mandel realized that sometimes purchasing enough tickets to cover every combination cost less than the potential jackpot. For instance, if the jackpot was $10 million and tickets cost $1, someone could buy all the tickets up and guarantee themselves a win, as well as a handsome profit. All he needed to do was raise enough capital to buy the tickets.

This is how he began forming lottery syndicates, where investors put in money to purchase lottery tickets, and the winnings were split. His first win using this system earned him a $19,000 jackpot, of which he pocketed $3,700. While that might not seem like much, it was enough to move his family out of Romania and settle down in Australia.

While in Australia, he won a staggering 12 jackpots, plus many other smaller prizes. Mandel set up larger and larger syndicates, all while staying within the rules and regulations of the game. At the time, it was legal to print tickets at home, which allowed Mandel to purchase millions of tickets and automate a way to fill in the different combinations.

Many people's routines would vastly improve if they got to hug a dog on their way to work or school. A boy named Ibrahim is leading by example, and in doing so, he has warmed the hearts of thousands around the globe. In a moving video posted on Facebook, the kid is seen stopping on his way to school to embrace a couple of stray dogs in Grozny, Chechnya.

The 30-second clip is a powerful statement of a boy's love for all canines. Filmed in 2021 by a resident in a nearby building, the footage begins with the boy giving a dog a hug, and petting the pooch's friend with a smile on his face. After walking away, Ibrahim seems to change his mind and quickly retraces his steps to give the first dog an even longer, tender embrace.

The sweetest aspect of this is that Ibrahim thinks no one is watching, and hes just being his truest, most loving self. It is unclear if this is the first time he encountered the dogs, or if he knows their story and believes they could use a hug on a chilly morning. Whatever the case, his love for the creatures is palpable.

For those who know Ibrahim, this is just another example of his goodness. I saw the video. Thats Ibrahim, Katerina, a friend of Ibrahims family, told The Dodo. He goes to school with my daughter and helps her carry her bag. Hes a very kind boy.

If there's one take away from Ibrahim's actions, it is that something as simple as a hug can brighten someone's dayand it goes both ways. You never know how many people you may be inspiring by simply sharing your love with the world.

France's highest administrative court ruled on June 29th that the countrys soccer federation has the right to ban players from wearing headscarves during matches, even if the action means limiting their freedom of expression.

The Council of State made this decision after a group of soccer players that wear religious headscarves, called Les Hijabeuses in French, referring to the hijab or religious headscarf in Islam, launched a campaign against the French Football Federation (FFF) and took legal action against their ban.

The court said that sports federations in France may impose on their players an obligation to wear neutral clothing during sporting competitions and events, in order to guarantee the smooth running of matches and prevent clashes or confrontation. It considers that the ban imposed by the FFF is appropriate and proportionate.

The ruling sparked a new debate on the principle of lacit in France, which is upheld by many French laws, such as Article 1 of the French Constitution and the 1905 French law on the Separation of the Churches and State. This principle is commonly interpreted as discouraging the participation of religion in government affairs and vice versa.

When I was growing up that was my parents mantra. That when I was an adult I was responsible for myself.

I started working when I was 15 and I attended a state college that I received a scholarship from. I graduated with only $7,500 debt and my job out of university came with a recruitment bonus that paid that off.

My brother didnt get the same talk. Our parents bought him a car when he was 16. He is smart and was accepted to a prestigious university. But no scholarship. And my parents earn enough that he does not qualify for financial aid.

I make a good living now and my parents and brother want me to help him out. The thing is he has a scholarship to a state school. He could just not go to an Ivy. But thats not good enough for them.

I reminded them what they always told me. They are mad that Im not willing to sacrifice my life to help another adult.

Stablecoin TrueUSD To Be Fully Controlled By Asian Owner "IndyWatch Feed Crypto"

In todays news, the prominent stablecoin TrueUSD with the ticker TUSD is now undergoing a management change.  According to a thread this morning by the projects official Twitter handle, Archblock Inc., the current TUSD operator, has begun the transfer of total control of Token to its Asian-based owners, Techteryx Ltd. 

Techteryx Finally Assumes Control Of TrueUSD

Back in December 2020, Techteryx acquired ownership of TUSD but hired Archblock to keep maintaining the stablecoins operations. And for the last two years in which Archblock remained TUSDs operator, Techteryx claims to have been focusing on expanding the tokens foreign use cases in the global markets.

However, Archblock has now commenced the transfer of control yesterday, July 13, marking the end phase of TUSDs international transition. Upon completion, Techteryx will reportedly assume full management of all aspects of the stablecoins operation.

These controls will include mining and redemptions, customer onboarding and compliance, conservation of fiat reserve, and maintenance of banking and fiduciary relationships.

During the transition period, Archblock will continue to support the US-based TUSD users, with Techteryx stepping in with the necessary guidance and further updates. 

Prior to todays news, TUSD has attracted some interest especially following Binances recent moves with the stablecoin. On June 21, the cryptocurrency exchange announced the launch of a TUSD zero-maker fee promotion for spot and margin trading pairs beginning from June 30.

Interestingly, Binance had minted $1 billion worth of TUSD on the Tron network a week before making that announcement becoming the largest holder of the token.

Why Ripples Victory Against The SEC May Be Short-Lived: Legal Expert "IndyWatch Feed Crypto"

The news of Ripples recent partial victory against the SEC after a legal battle spanning almost three years sent a ripple of joy around the entire crypto industry. However, according to a legal expert on Twitter, Lawyer Bryan Jacoutot, the victory might be short-lived as SEC has enough grounds to appeal the decision and drag this thing out for a lot longer.

The SECs Lawsuit Against Ripple Labs

The SEC filed a lawsuit against Ripple Labs in December 2020, alleging that Ripple had conducted an unregistered securities offering worth over $1.3 billion through the sale of XRP. According to the SEC, XRP is a security under federal securities laws. But the court determined on June 13 that the random programmatic sale of XRP to regular investors does not constitute the sale of an unregistered security under Howey. 

However, sales to institutional investors fall under Howey, which is used in the United States to determine whether a transaction qualifies as an investment contract. In this case, the Court found that the buyers couldnt know who was selling them the XRP, unlike the institutional investors who would expect Ripple Labs to use the capital for the betterment of the Ripple ecosystem.

According to Jacoutot, the Courts reasoning is weak and Howey was misapplied in the case. The Court reasoned that regular investors bought XRP fully knowing that it is subject to the general cryptocurrency market trends, especially secondary sales of XRP tokens. However, Jacoutot believes those buying XRP would have also expected to make a profit from the efforts of Ripple Labs.

The attorney also made a case of the Ethereum Foundation, as everyone who took part in the pre-sale of ETH knew they were buying from Ethereum Foundation. When looking at the XRP ruling in a similar manner, this would mean institutional investors of the ETH presale also bought unregistered securities.

What Does The Ruling Mean?

According to Jacoutot, the ruling opens up a few loopholes that can be exploited. In a tweet by attorney Joe Carlasare on Twitter, it explains that the logic of the ruling leaves an opening that can be used to lawfully launch a pyramid scheme. In this case, profits from the programmatic sales to retail investors can be distributed to institutional investors.

The last straw last time was when one of the little ones had a medical emergency so we took her to the ER directly from school and asked her to watch the younger one since the babysitter had to leave. Ark Invest Sells Over $50 Million Worth of Coinbase Shares Amidst Stock Rally "IndyWatch Feed Crypto"

Cathie Woods investment firm, Ark Invest, has made significant moves in its holdings of Coinbase shares, selling over $50 million worth of shares as the cryptocurrency exchanges stock continues to surge. 

This marked the second time in a week that Ark Invest reduced its stake in Coinbase, reflecting its active management approach amid a backdrop of regulatory developments and industry optimism.

At the same time, Ark Invest has been actively investing in other notable companies, including Meta Platforms and Robinhood. 

Ark Invest Cashes In on Coinbase Rally

Ark Invest, led by Cathie Wood, sold a total of 478,356 shares of Coinbase on Friday, amounting to more than $50 million. The sales were spread across Arks flagship fund, Ark Innovation ETF, which sold 263,247 shares, Ark Next Generation Internet ETF, which sold 93,227 shares, and Ark Fintech Innovation ETF, which offloaded 121,882 shares. 

This decision comes on the heels of Coinbases role as a surveillance-sharing partner for several spot Bitcoin ETF applicants, including industry giants BlackRock and Fidelity. Furthermore, recent legal rulings surrounding the status of the cryptocurrency XRP have added to the overall industry optimism.

However, despite the sales, Ark Invest remains the second-largest owner of Coinbase shares, holding a 6.30% stake.

Looking To The Future With Meta Platforms and Robinhood

While reducing its Coinbase holdings, Ark Invest has also been actively investing in other crypto-adjacent companies. The firm initiated purchases of shares in Meta Platforms (formerly Facebook) and Robinhood. In June, Ark Innovation ETF acquired 69,793 Meta shares, while Ark Fintech Innovation ETF purchased 111,843 shares of Robinhood. 

Additionally, the Ark Next Generation Internet ETF increased its holdings with 12,559 Meta shares and 169,116 Robinhood shares. These strategic investments reflect Ark Invests ongoing strategy to navigate the evolving digital asset market.

I (22F) have two brothers for the sake of this post Ill call them John (18M) and Terry (16M).

My mom (50F) has always had a stronger liking towards both John and Terry, ever since they were little they could do no harm in her eyes.

Since I was little I would always get in trouble for everything John and Terry did, if they hurt each other it was my fault, if they did something wrong it was my fault etc. My mom always raised me as a caregiver, I had to basically take care of my little brothers, I was the one cooking, cleaning, washing and ironing their clothes, buying them stuff they needed, helping them with their homework etc.

However, due to me always doing everything for them, they have become the most useless human beings on the entire earth. They are almost adults and still wont do their own laundry, wont help with the cooking, wont clean anything that they made a mess and are just overall grubs, leaving their dirty tissues and clothes everywhere.

In popular culture, Japan is home to some very large creatures like Godzilla and Mothra. If you visit the Diorama Restaurant in Osaka, Japan, you will find giant cats as well. The charming eatery, which doubles as a cat shelter, is home to beautiful scale models of landscapes and tiny towns which appear to be overrun by large felines. As if that weren't enticing enough, the restaurant also features a miniature train that goes around the locale, although its rides are rarely free of incidents. A small camera attached to the train captures the many ways these trips are interrupted by seemingly monumental kitties.

It is incredibly entertaining to see the (very-normal-sized) cats interact with the miniature train as it travels around the grassy hills of the restaurant. The true magic of the experience, however, lies in seeing the point-of-view footage of these epic rides. Sometimes, the train bumps into a sleepy cat who, despite seeing the convoy coming in, doesn't want to move from its snuggly spot. Other times, a feline silently prowls the train until an off-camera force turns it on its side. Since every cat's personality differs, it adds a layer of adrenaline to each trip.

Due to the nature of the place, the Diorama Restaurant is reservation-only. The Diorama Cafeteria is open in three parts, with lunchtime, tea time, and time spent together, reads their website. There is no difference in the service contents except for the contents of the meal each time. Occasionally, a free slot will open up, so you can check their Twitter to snag a same-day reservation.

Sound of Freedom Is Hard to Watch. Heres Why We Need to See It Anyway. "IndyWatch Feed Survival"

The movie Sound of Freedom was both one of the best and worst movies Ive ever seen.  Best because the story was interesting, the protagonists were likable and engaging, and the plot moved quickly. But the worst part is that it was also very hard to watch because I went into the theater knowing that the movie is largely true.  

The movie starts off with the kidnapping of a brother and sister from Honduras by a young woman posing as a talent agent.  You then see the American Homeland Security agent, Tim Ballard, recover the little boy.  After the little boys rescue, he asks Tim if he can find his sister, too, and most of the movie involves Tim trying to find out what happened to the little girl.  

Its an action story of a man trying to put a family back together; its also a story of Tim Ballards personal journey in leaving the security of a government job to do what he believes is right.

Heres some background.

I watched Jordan Petersons interview with both Tim Ballard and Jim Caviezel, who plays Tim in the movie, and I recommend it because it adds a bit of depth to what was going through Tims mind.  Tims wife is only a very minor character in the movie, but in his interview with Jordan Peterson, Tim tells Jordan that she was actually one of the major drivers behind his actions.

"IndyWatch Feed Craft"

So, I was supposed to be going to Medeltisveckans on Gotland in Sweden this August.  But Im not.  And thats OK (well, the not going.  Not how it happened.  That wasnt OK.  And Im dealing with that).

But the good news is, Im going to be doing something even better.  Youll hear about that soon

But, I spent 4 months planning for Medeltisveckans, which obviously means sewing!  Medieval sewing.

My big worry about Medeltisveckans is that by all accounts is usually hot.  So I needed to plan a capsule wardrobe of >1 suitcase worth of Medieval clothes that I wouldnt overheat in, and that would be as light as possible (because, luggage allowances).

My solution?  My yellow 1370s dress, and one of the short sleeved dresses from the famous Herjolfsnes finds in wool, with the option to wear it with or without pin on sleeves and alone or over the yellow 1370s dress so many different options, and the looseness of the Herjolfsnes dress would make it cool and breezy in the heat.

So, I patterned up Herjolfsnes 39 from Medieval Garments Reconstructed (its on page 58, and featured on the front cover)

This was back in the late 1980s, shortly after I dropped everything and went to live with her. Ill never stop dwelling on that moment and the system that failed my sister. Considerate Tall Guy at Show Offers to Narrate On-Stage Action to Short Woman Behind Him "IndyWatch Feed Satire"

Every time I tried to contort my body to get a good view of the band, this guy did too. Almost instinctively, said Belanger. Not to mention his narration skills were complete dog shit. He just kept naming the song the band was playing in real time as if he thought blocking my view meant obstructing the sound of their music. Cant believe this was the show I forgot to bring my trusty little step stool I otherwise never leave home without.

Music expert Chase Clay outlined some basic manners showgoers need to be aware of.

Audiences usually have to adhere to some sort of etiquette during live music, especially if youre a beanstalk of a human, said Clay. So if youre tall and you find yourself in this situation, you can offer to hoist short people up on your shoulders so they can see, or you can lift them up in the air Dirty Dancing-style. Or better yet, you can do everyone a favor and leave the show entirely to let the normal-sized people enjoy the action for once, you freak of nature. Otherwise, just make sure to have fun.

We had to start rationing the lidocaine patches, and I cant take two steps without tripping over a guy laying on the floor because he threw his back out and all they want to talk about is some guy named Andrew Allen. If we get any more sprained ankles today were going to have to start tearing towels to make wraps.

Local rent-a-cops tasked with patrolling the citys parking lots admit they do not share the same enthusiasm as the skaters.

Im sick and tired of slowly chasing these damned punk middle-aged skaters off the lot, said security guard Glenn Crosby, scratching a local brewery sticker off a railing. They leave their empty ibuprofen bottles everywhere, and even when theyre not skating they just loiter here for hours, icing their knees. These guys need to grow up and go hang out at a Home Depot like normal middle-aged guys.

If Home Ownership Is So Elusive, Why Did I Inherit Three? "IndyWatch Feed Satire"

Not a day goes by that I dont hear someone bitching about not being able to afford a home. They claim the economy is dogshit and that their crippling student debt, combined with high interest rates, has killed any chance of them ever purchasing property. Real sad face emoji-type stuff. But if home ownership is so difficult to achieve, how the heck did I successfully inherit three?

I think most Gen Zers and Millennials who dont have homes only have themselves to blame. They waste their money on luxuries like rent instead of being fiscally responsible and using the money they inherit when their wealthy relatives croak on down payments.

Just the other day I overheard a bunch of college grads at a car wash bemoaning their economic reality, complaining about needing multiple precarious service jobs just to survive. I wish I had a chance to give them some real estate advice but they finished detailing my Land Rover pretty quickly and I was in a bit of a hurry.

Dont get me wrong, I do acknowledge that there are real challenges when trying to secure your first home. I, for example, had an uphill battle to even get the keys to any of the three houses Gramps left me because my butthurt siblings who were cut out of the will took me to court. The anxiety I suffered during that unbearable two-week period of housing insecurity is something Ill never let go.

At the end of the day, I think this generation should take a page from the bestselling book, The Secret, and start manifesting a positive mindset to achieve their housing dreams, including owning three palatial estates by the ocean, like me. And if that doesnt work, theres always insurance fraud. All it takes is a little accidental fall in the lobby of a five-star hotel and you could very well be on your way to finally owning your first property.

When I (17 M) was 8, my parents bought me a piano and signed me up for lessons. I was super excited because I love music. Over time I kind of became known as the piano guy at school. I play at school concerts, accompany the school jazz choir, and play once a week for the residents at a couple of retirement homes in our town.

When I was 15, I started to talk about quitting lessons, and my parents quickly tried to guilt me out of it. I told them I wanted to try other things, and that between piano and studying, I didnt have much time left for other extra curriculars. My Dad proposed a deal.

A jury in New Mexico awarded $485 million to an 8-year-old female victim of Clarence Garcia, a 66-year-old foster parent in New Mexico who sexually abused the girl while she was in his care.

A Rio Arriba County jury awarded an astounding $485 million in a lawsuit. It was filed on behalf of a girl who was repeatedly raped by her foster father, Clarence Garcia.

I think the message that was sent first and foremost is that this little girl matters, said Joshua Conaway, an attorney in the case.

What Is the Cell Danger Response? "IndyWatch Feed Health"

Since early 2020, I have worked with patients with spike protein injuries, either from COVID-19 or the vaccines. Now and then, I've observed a specific treatment for a spike protein injury have a rapid effect that was so dramatic it would have been difficult to believe it had happened had I not witnessed it firsthand.

Whenever I spotted a treatment doing that, I asked, "Why did this happen?" Over time, I realized two mechanisms appeared to be able to account for almost every case where I observed this happen.

The first was that impaired fluid circulation in the body was restored, most commonly by restoring the physiologic zeta potential (something the spike protein is uniquely suited to inhibit). Since this is a complex but relatively unknown topic, I've worked to explain what zeta potential is, how its disruption creates illness by impairing fluid circulation throughout the body, and the methods I know of which can restore it.

Note: In parallel to doing this, I also discussed the fourth phase of water (something also critical for the health of the body) as the two concepts are deeply interrelated.

The second was that the Cell Danger Response (CDR) was deactivated. Since there is much more awareness about this (still relatively unknown) concept in the integrative medical field, I felt zeta potential needed to be covered first. Nonetheless, the CDR is an essential concept to understand, and like zeta potential, it plays a foundational role in explaining and addressing many of the complex conditions we face today.

Before I go further, I would like to acknowledge two of my colleagues who have a great deal of experience working with the CDR that assisted me in drafting this series (and both independently observed that the COVID vaccines triggered the CDR).

What Is the Cell Danger Response?

For cells to survive, something has to protect them from the innumerable threats they encounter. In complex organisms, we typically assign that role to the immune system. How to Boost Brain Health and Reverse Neurological Disease "IndyWatch Feed Health"

Your brain is a really important part of your body. I'm sure no one would disagree with that. In this interview, Dr. Lee Cowden, co-founder of the Academy of Comprehensive Integrative Medicine (ACIM), discusses some really simple and inexpensive strategies that boost brain health and support neuroregeneration. This is also the topic of this year's ACIM conference, which will be held in Orlando, Florida, in November.

The Academy of Comprehensive Integrative Medicine was founded in 2008 with the vision of educating integrative practitioners around the globe, for the most part online. "Many practitioners are so busy that they can never take the time to go to all of the meetings they need to go to, to learn what they need to learn," Cowden says, "so we decided to start putting educational courses online."

ACIM Offers Training Programs

In 2014, ACIM began offering a 300-hour online integrative medicine fellowship training program. Workshops are held in Dallas every few months to allow practitioners to get hands-on training. In 2018, there were hands-on conferences in July and September 20. Those were all preludes to the main conference in Orlando in November.

"Everything we're doing this year is focused on the brain [and] neuroregeneration," Cowden says. "With integrative medicine, we look at the causation, and see if we can resolve the causation of the illness. Once the cause goes away, usually the illness goes away."

You don't have to be a health care professional to benefit from ACIM. Even laypeople can study with ACIM.

The academy offers a core wellness coach program that includes 70 hours of training. "I think every integrative practitioner needs to have half a dozen or more wellness coaches working with them to hold the hands of the patients until they get well," Cowden says.

ACIM Conference 2018: Neuroregeneration

The November conference focused on strategies that can help regenerate all types of neurological conditions, including Alzheimer's, autism, multiple sclerosis, amyotrophic lateral sclerosis, Parkinson's disease and other less severe conditions such as the neurological effects of fibromyalgia, and less common conditions such as reflex sympathetic dystrophy and peripheral neuropathy.

Abstract:  Elevated inflammation has been associated with adverse mood states, such as depression and anxiety, and antioxidant nutrients, such as vitamin C, have been associated with decreased inflammation and improved mood. In the current study comprising a cohort of pregnant women with depression and anxiety, we hypothesised that elevated inflammation would be associated with adverse mood states and inversely associated with vitamin C status and that multinutrient supplementation would optimise vitamin concentrations and attenuate inflammation. Sixty-one participants from the NUTRIMUM trial had blood samples collected between 12 and 24 weeks gestation (baseline) and following 12 weeks of daily supplementation with a multinutrient formula containing 600 mg of vitamin C or active placebo. The samples were analysed for inflammatory biomarkers (C-reactive protein (CRP) and cytokines) and vitamin C content and were related to scales of depression and anxiety. Positive correlations were observed between interleukin-6 (IL-6) and all of the mood scales administered (0.05). Supplementation with the multinutrient formula resulted in a significant increase in the vitamin C status of the cohort (= 0.007) but did not affect the inflammatory biomarker concentrations (>0.05). In conclusion, greater systemic inflammation was associated with worse mood states; however, 12-week multinutrient supplementation did not alter inflammatory biomarker concentrations. Nevertheless, the vitamin C status of the cohort was improved with supplementation, which may aid pregnancy and infant outcomes.

VC and VDshowed similar beneficial effects as mentioned above in HFD-induced obese mice. Interestingly, VC and VDreshaped the gut microbiota composition; improved gut barrier integrity; ameliorated oxidative stress and inflammation in the gut-liver axis; inhibited bile acid salt reflux-related ASBT; activated bile acid synthesis-related CYP7A1, bile acid receptor FXR, and bile acid transportation-related BSEP in the gut-liver axis; and improved bile secretion, thus decreasing the expression of FAS in the liver and efficiently ameliorating MAFLD in mice.Together, the results indicate that the anti-MAFLD activities of VC and VDare linked to improved gut-liver interactionsregulation of the gut microbiota and bile acid metabolism, and they may therefore prove useful in treating MAFLD clinically.

Abstract:  BACKGROUND: This meta-analysis aimed at investigating the pooled evidence regarding the effects of intravenous vitamin C (IVVC) on mortality rate in critically ill patients.METHODS: Databases including Medline, Embase, and Cochrane Library were searched from inception to October, 2022 to identify RCTs. The primary outcome was the risk of overall mortality. Subgroup analyses were performed based on IVVC dosage (i.e., cut-off value: 100 mg/kg/day or 10000 mg/day). Trial sequential analysis (TSA) was used to examine the robustness of evidence.RESULTS: A total of 12 trials including 1,712 patients were analyzed. Although meta-analysis demonstrated a lower risk of mortality in patients with IVVC treatment compared to those without [risk ratio (RR): 0.76, 95% CI: 0.6 to 0.97,= 0.02,= 36%, 1,711 patients), TSA suggested the need for more studies for verification. Moreover, subgroup analyses revealed a reduced mortality risk associated with a low IVVC dosage (RR = 0.72,= 0.03, 546 patients), while no beneficial effect was noted with high IVVC dosage (RR = 0.74,= 0.13,= 60%, 1,165 patients). The durations of vasopressor [mean difference (MD): -37.75 h, 404 patients) and mechanical ventilation (MD: -47.29 h, 388 patients) use were shorter in the IVVC group than those in the controls, while there was no significant difference in other prognostic outcomes (e.g., length of stay in intensive care unit/hospital) between the two groups.CONCLUSION: Although intravenous vitamin C as a monotherapy reduced pooled mortality, durations of vasopressor use and mechanical ventilation, further research is required to support our findings and to identify the optimal dosage of vitamin C in the critical care setting.SYSTEMATIC REVIEW REGISTRATION: https://www.crd.york.ac.uk/prospero/, identifier CRD42022371090.

Moreover, the creation of ketone bodies may improve brain neuronal function, decrease inflammatory expression and reactive oxygen species (ROS) production, and restore neuronal metabolism. As a result, the KD has drawn attention as a potential treatment for neurological diseases, such as T2DM-induced dementia. This review aims to examine the role of the KD in the prevention of dementia risk in T2DM patients and to outline specific aspects of the neuroprotective effects of the KD, providing a rationale for the implementation of dietary interventions as a therapeutic strategy for T2DM-induced dementia in the future.

Abstract:  Bipolar affective disorder (BPAD) is a chronic mental disorder, characterised by mood swings, alternating between depression and manic or hypomanic episodes. Unfortunately, in some patients pharmacological treatment does not bring satisfactory results, and a certain group of patients shows resistance to treatment. Therefore, other treatment methods are sought after, including a change in diet. The most promising nutrition model is the ketogenic diet. In the presented case study of a male patient, thanks to the introduction of the ketogenic diet, full remission of the disease was achieved, doses of lamotrigine were reduced and quetiapine was completely discontinued. Previously, neither lamotrigine monotherapy nor combined treatment with quetiapine achieved euthymia. The effects of the diet may be related to, among others, the influence on ionic channels and increase in blood acidity (similarly to mood stabilisers), increase in gamma-aminobutyric acid (GABA) concentration, modulation of GABAA receptors and blocking of AMPA receptors by medium-chain fatty acids. The ketogenic diet influences glutamate metabolism and nerve cell metabolism, which uses ketone bodies as energy sources. Ketosis can also stimulate the biogenesis of mitochondria, improve brain metabolism, act as a neuroprotective factor, as well as increase glutathione synthesis and reduce oxidative stress. However, there is a need for carefully planned studies, with an appropriate representative group, to verify the potential benefits and risks of introducing the ketogenic diet in patients with BPAD.

Abstract:  OBJECTIVE: The ketogenic diet (KD) is one of the main treatments for drug-resistant epilepsy. However, there have been few multicenter reports on the use of the KD for the treatment of Dravet syndrome (DS). The aim of this study was to analyze the efficacy and safety of this approach based on a large number of multicenter cases.METHODS: This was a retrospective, multicenter cohort study from 14 centers in China. All patients were treated with the KD. We compared the effects of KD intervention time, age, and other factors.RESULTS: From March 2014 to March 2020, we treated 114 patients with DS with the KD. The male-to-female ratio was 67:47. The KD median initiation age was 3 y and 4 mo, and the median number of antiseizure medications (ASMs) was 2.4. KD therapy was the first choice for three patients. Exactly 10.5% of the patients started KD therapy after failure of the first ASM therapy, with 35.1% after failure of the second, 44.7% after the third, and 7% after the fourth or more. After KD therapy for 1, 3, 6, and 12 mo, the seizure-free rates were 14%, 32.5%, 30.7%, and 19.3%, respectively; KD efficacy (50% reduction in seizure frequency) were 57.9%, 76.3%, 59.6%, and 43%, respectively; the retention rates were 97.4%, 93%, 71.9%, and 46.5%, respectively; and the rates of adverse events were 25.2%, 19.9%, 11%, and 5.7%, respectively.CONCLUSIONS: Real-world, multicenter data analysis showed that the KD is effective for patients with DS and has a low incidence of side effects.

A jury in New Mexico awarded $485 million to an 8-year-old female victim of Clarence Garcia, a 66-year-old foster parent in New Mexico who sexually abused the girl while she was in his care.

A Rio Arriba County jury awarded an astounding $485 million in a lawsuit. It was filed on behalf of a girl who was repeatedly raped by her foster father, Clarence Garcia.

I think the message that was sent first and foremost is that this little girl matters, said Joshua Conaway, an attorney in the case.

And even though California has spent a whopping 17.5 billion dollars on homelessness during the last four years, the homeless population in the state has just continued to grow

Abstract:  The role of pescadillo1 (PES1) in regulating vascular permeability has been unknown. This study probes the role of PES1 and its mediated molecular mechanism in modulating vascular hyperpermeability in diabetic mice. Male C57BL/6J and db/db mice were fed a standard diet and a ketogenic diet (KD). Meanwhile, mouse vascular endothelial cells (MVECs) were treated with-hydroxybutyric acid (-HB), Pes1 siRNA or a Pes1 overexpression plasmid. Additionally, knockout (KO) of Pes1 in mice was applied. After 12weeks of feedings, enhanced vascular PES1 expression in diabetic mice was inhibited by the KD. The suppression of PES1 was also observed in-HB-treated MVECs. In mice with Pes1 KO, the levels of vascular VEGF and PES1 were attenuated, while the levels of vascular VE-cadherin, Ang-1 and Occludin were upregulated. Similar outcomes also occurred after the knockdown of Pes1 in cultured MVECs, which were opposite to the effects induced by PES1 overexpression in MVECs. In vitro and in vivo experiments showed that high glucose concentration-induced increases in vascular paracellular permeability declined after MVECs were treated by-HB or by knockdown of Pes1. In contrast, increases in vascular permeability were induced by overexpression of Pes1, which were suppressed by coadministration of-HB in cultured endothelial cells. Similarly declines in vascular permeability were found by Pes1 knockdown in diabetic mice. Mechanistically,-HB decreased PES1-facilitated ubiquitination of VE-cadherin. The KD suppressed the diabetes-induced increase in PES1, which may result in vascular hyperpermeability through ubiquitination of VE-cadherin in type 2 diabetic mice.

KD-fed mice show reduced brain edema by decreasing aquaporin-4 (AQP4) content and maintaining its polarized expression. 16S rRNA gene amplicon sequencing results show that KD reduces the Chao 1 index of gut microbiota-diversity, and significant separation is detected in the-diversity analysis between the control and KD-fed mice. KD increases the relative abundance of Firmicutes and Proteobacteria and decreases that of Bacteroidetes. Hypoglycemia can reduce SOD and GSH-PX levels while increasing TNF-, IL-1, and IL-6 mRNA levels in the brain tissues of mice. KD alleviates hypoglycemia-induced neuroinflammation by inhibiting microglia activation and TLR4/p38MAPK/NF-B signaling pathway. Importantly, antibiotic cocktail depletion of the gut microbiota weakens anti-inflammatory and antioxidation responses in KD-fed mice.CONCLUSION: Collectively, these findings suggest that KD alleviates hypoglycemia-induced brain injury via gut microbiota modulation, which may provide novel insights into the therapy for hypoglycemia.

The review provides an update on clinical and preclinical research on the effects of KD or medium-chain triglyceride supplementation on symptoms and pathophysiology in AD. We also detail the potential mechanisms of KD, involving amyloid and tau proteins, neuroinflammation, gut microbiota, oxidative stress, and brain metabolism. We aimed to determine the function of the KD in AD and outline important aspects of the mechanism, providing a reference for the implementation of the KD as a potential therapeutic strategy for AD.

Histological analysis revealed that KD alleviated structural destruction and fibrosis in obstructed kidneys and reduced the expression of the fibrosis protein markers-SMA, Col1a1, and Col3a1. Expression of the rate-limiting enzymes involved in fatty acid oxidation (FAO), Cpt1a and Acox1, significantly decreased after UUO and were upregulated by KD. However, the protective effect of KD was abolished by etomoxir (a Cpt1a inhibitor). Besides, our study observed that KD significantly suppressed UUO-induced macrophage infiltration and the expression of IL-6 in the obstructive kidneys. In NRK52E cells, fibrosis-related signaling was increased by TGF1 and reduced by-OHB.-OHB treatment restored the impaired expression of Cpt1a. The effect of-OHB was blocked by siRNA targeting free fatty acid receptor 3 (FFAR3), suggesting that-OHB might function through the FFAR3-dependent pathway.DISCUSSION: Our results highlight that KD attenuates UUO-induced renal fibrosis by enhancing FAO via the FFAR3-dependent pathway, which provides a promising dietary therapy for renal fibrosis.

This study aimed to determine whether KD protects against renal fibrosis in unilateral ureteral obstruction (UUO) models and the possible mechanisms. The ketogenic diet, according to our findings, reduces UUO-induced kidney injury and fibrosis in mice. KD dramatically decreased the number of F4/80macrophages in kidneys. Next, immunofluorescence results revealed a reduction in the number of F4/80Ki67macrophages in the KD group. Furthermore, our study evaluated the impact of-hydroxybutyric acid (-OHB) in RAW246.7 macrophages in vitro. We found that-OHB inhibits macrophage proliferation. Mechanistically,-OHB inhibits macrophage proliferation may be via the FFAR3-AKT pathway. Collectively, our study indicated that KD ameliorates UUO-induced renal fibrosis by regulating macrophage proliferation. Abstract:  OBJECTIVE: The aim of this study isto assess the efficacy of a very low-calorie ketogenic diet (VLCKD) method vs a Mediterranean low-calorie diet (LCD) in obese polycystic ovary syndrome (PCOS) women of a reproductive age.DESIGN: Randomized controlled open-label trial was performed in this study. The treatment period was 16 weeks; VLCKD for 8 weeks then LCD for 8 weeks, according to the Pronokalmethod (experimental group; n = 15) vs Mediterranean LCD for 16 weeks (control group; n = 15). Ovulation monitoring was carried out at baseline and after 16 weeks, while a clinical exam, bioelectrical impedance analysis (BIA), anthropometry, and biochemical analyses were performed at baseline, at week 8, and at week 16.RESULTS: BMI decreased significantly in both groups and to a major extent in the experimental group (-13.7% vs -5.1%, P = 0.0003). Significant differences between the experimental and the control groups were also observed in the reduction of waist circumference (-11.4% vs -2.9%), BIA-measured body fat (-24.0% vs -8.1%), and free testosterone (-30.4% vs -12.6%) after 16 weeks (P = 0.0008, P = 0.0176, and P = 0.0009, respectively). Homeostatic model assessment for insulin resistance significantly decreased only in the experimental group (P = 0.0238) but without significant differences with respect to the control group (-23% vs -13.2%, P>0.05). At baseline, 38.5% of participants in the experimental group and 14.3% of participants in the control group had ovulation, which increased to 84.6% (P = 0.031) and 35.7% (P>0.05) at the end of the study, respectively.CONCLUSION: In obese PCOS patients, 16 weeks of VLCKD protocol with the Pronokalmethod was more effective than Mediterranean LCD in reducing total and visceral fat, and in ameliorating hyperandrogenism and ovulatory dysfunction.SIGNIFICANCE STATEMENTS: To the best of our knowledge, this is the first randomized controlled trial on the use of the VLCKD method in obese PCOS. It demonstrates the superiority of VLCKD with respect to Mediterranean LCD in reducing BMI with an almost selective reduction of fat mass and a unique effect of VLCKD in reducing visceral adiposity, insulin resistance, and in increasing SHBG with a consequent reduction of free testosterone. Abstract:  Studies have shown ketogenic diets (KD) started from early middle-age improved health span and longevity in mice. KDs started later in life or administered intermittently may be more feasible and promote compliance. Therefore, this study sought to test if continuous or intermittent KDs started in late-middle-aged mice would improve cognition and motor function at advanced age. Eighteen-month-old male C57BL/6JN mice were assigned to an isocaloric control (CD), KD, or intermittent ketogenic (IKD, 3-day KD/week) diet. A panel of behavior tests were performed to assess cognitive and motor functions with aging. Y-maze alternation rate was higher for both IKD and KD mice at 23 months of age and for KD mice at 26 months indicating an improved spatial working memory. Twenty-six-month-old KD mice also showed better spatial learning memory in Barnes maze when compared to the CD. Improved grid wire hang performance was observed in aged IKD and KD versus CD mice indicating better muscle endurance under isometric contraction. A reduced level of circulating proinflammatory cytokines in aged KD (IL-6 and TNF-) and IKD (IL-6) mice may contribute to the phenotypic improvements observed with these interventions. This study demonstrates that when initiated at late-middle age, the KD improved measures of spatial memory and grid wire hang performance in aged male mice, with IKD showing results intermediate to the CD and KD groups.

Over the four months on the ketogenic diet, the patient reported losing 20 kg of weight, reduction in blood pressure (without antihypertensives), and complete alleviation of asthma symptoms. This case report is important as the control of asthma after a ketogenic diet is not studied well in humans and therefore needs to be studied extensively.

Beta-hydroxybutyrate (BHB), the major ketone body produced during the carbohydrate deprivation that occurs in KD, is assumed to have neuroprotective effects, although the molecular mechanisms responsible for these effects are still unclear. Microglial cell activation plays a key role in the development of neurodegenerative diseases, resulting in the production of several proinflammatory secondary metabolites. The following study aimed to investigate the mechanisms by which BHB determines the activation processes of BV2 microglial cells, such as polarization, cell migration and expression of pro- and anti-inflammatory cytokines, in the absence or in the presence of lipopolysaccharide (LPS) as a proinflammatory stimulus. The results showed that BHB has a neuroprotective effect in BV2 cells, inducing both microglial polarization towards an M2 anti-inflammatory phenotype and reducing migratory capacity following LPS stimulation. Furthermore, BHB significantly reduced expression levels of the proinflammatory cytokine IL-17 and increased levels of the anti-inflammatory cytokine IL-10. From this study, it can be concluded that BHB, and consequently the KD, has a fundamental role in neuroprotection and prevention in neurodegenerative diseases, presenting new therapeutic targets.
