The American beaver is a mighty creature. Once incredibly
numerous, they figured heavily in indigenous worldviews in the Americas. When
Europeans arrived, they prized the beaver for its pelt, and it was
hunted almost to extinction over the following centuries. However,
these small mammals have been part of another enduring legacy
through their effect on the changing American landscape. In fact,
one enthusiast recorded the dams created by beavers in the mid-19th
centuryand nearly 75% of those he noted are still visible
today.
Lewis Henry Morgan was a railroad magnate and politician with a
passion for anthropology and natural sciences. Michigan's Upper
Peninsula turned out to be fascinating to him as he planned his
railroad through the state. Throughout the 1850s and 1860s, he
observed the beaver, (apparently) his new favorite creature.
This observation yielded an impressive 396-page book
impressively entitled The American Beaver and His
Works. Within each folio was tucked a map of the
Ishpeming, Michigan, area. Sixty-four beaver dams dot the
landscape, interrupting streams and creating ponds. To create such
a map, Morgan used his own naturalist skills combined with the
drawing techniques of his railroad engineers. It was published in
1868.
So what have the busy beavers been up to in Michigan in the past
century and a half? As it turns out, largely maintaining these
historic dams. Despite the loss of habitat and population, 46, or
75%, of the dams Morgan noted are still visible on the landscape.

New York-based artist Xiao Wang masterfully sets the
mood in his portraits with uncanny environments. Although
realistically rendered, the color palettes have a slightly
distorted quality that speaks to what each figure is feeling or
thinking at the moment. From swirling violet clouds to blue-toned
leaves, these backgrounds help us understand what Wang's subjects
are trying to saybut cannot.
Originally from China, Wang's experience as an immigrant has a
major influence on his portraits. The work describes the feeling of
being trapped between two worlds, navigating feelings of
uncertainty and unease while trying to assimilate. His subjects
wear slightly enigmatic expressions that make it hard to discern
exactly what they are thinking. While some stare off into the
distance, others even look like they are smiling. The cues to what
is truly going on in their head exist in the settings.
Most of the figures depicted in Wang's work have a personal
connection to the artist, either as a friend, partner, or Wang
himself. The juxtaposition of a naturalistic approach to human
anatomy and the altered colors makes it seem like these people
exist somewhere between reality and a dream. And it is in this
place that we can better understand their state of mind.

next month by a leading global health body, according to two

UK-based artist Lisa Stevens pushes the boundaries
of ceramics with her aquatic-inspired
creations. Embedded with intricate details, these pieces mimic
the colors and forms found by the sea. Featuring bright flowers and
textured ridges, any of these sculptures look like they could've
washed up on shore.
Stevens has been capturing natural oceanic motifs for years.
While her art has become increasingly elaborate, her process
remains the same. She begins each piece from clay and uses various
tools to sculpt and shape the material into the desired form. This
can mean adding tiny indentations all along the surface, scratching
long lines, and crafting flower petals. Although this process can
be labor-intensive, it results in a very tactile finish with an
eclectic mix of textures.
Once the building is done, Stevens applies a carefully chosen
array of underglazes and stoneware glazes to the
sculpturessometimes even adding melted glassto give the pieces a
more aquatic feeling. Her palette usually includes blues and greens

Bitcoin needs to change! In the realm of Bitcoin heresy, before
you prepare to hang me from the gallows for that statement, permit
me to express my final thoughts. Allow me to provide a brief
introduction I am not a galaxy-brained, shadowy super coder. I must
admit to my relative ignorance in the fields of computer science
and cryptography. I do not work within the Bitcoin industry.
Instead, I am an ordinary pleb, toiling away in construction to
make ends meet, striving to maintain humility and gain a
comprehensive understanding of the true nature of Bitcoin. So, why
do I say that Bitcoin must change?
Let us commence by reflecting upon the adage, "Not your keys,
not your coins," which resounds as one of the loudest mantras in
the Bitcoin community, and rightfully so. When your private keys
are held by a centralized exchange or custodian on your behalf, you
effectively surrender control over your funds. This situation
introduces counterparty risk, as you become reliant on the security
practices and integrity of the third party entrusted with your
keys.
The philosophy underlying "not your keys, not your coins" aligns
with the broader principles of decentralization and censorship
resistance. These principles seek to empower individuals with
financial sovereignty and eliminate the necessity for
intermediaries. It underscores the significance of personal
responsibility, security and self-reliance in this digital era
where governments increasingly employ technology as a tool for
controlling individuals.
Now, you may ask, what is the issue at hand? Well, the truth of
the matter is that, under the current implementation of the Bitcoin
protocol, self custody does not scale. Many Bitcoin enthusiasts
choose not to critically analyze this reality. However, we cannot

The US Department of Labor stated that an employee testified in
court that Che Garibaldi, owner of Taqueria Garibaldi, which
operates two branches in California, offered a person who
identified as a priest to hear their confessions during work
hours.
"The priest urged workers to 'get their sins out,' and asked
employees if they had stolen from the employer, been late for work,
had done anything to harm their employer or if they had bad
intentions toward their employer," a statement from the US
Department of Labor said.

by Garibaldi.
Taqueria Garibaldi and its owners and operators did not respond to
requests for comment. In response to the incident, a spokesperson

connection" with the alleged priest.
Authorities uncovered more findings during their investigation
into the restaurant and its owner. They found that Taqueria
Garibaldi denied employees overtime pay while managers illegally
received bonuses from the employee tip pool. Investigators also
found that the restaurant threatened to retaliate against its
employees and "adverse immigration consequences" for

Six months after a far-right Swedish-Danish activist and
politician enraged the entire Muslim community by publicly burning
the Quran in front of the Turkish embassy in Swedens capital,
Stockholm, another Quran-burning stunt occurred in the capital,
this time during the celebration of Eid al-Adha, or the Feast of
Sacrifice.
The person responsible for this latest Quran-burning publicity
stunt was not a far-right provocateur like Rasmus Paludan but a
refugee from Iraq identified as Salwan Momika. He performed the act
outside the Stockholm Central Mosque on June 28th while Muslims
were celebrating the Feast of Sacrifice, which commemorates the
willingness of Abraham to follow Gods command by attempting to
sacrifice his son.
Momika began the stunt by walking near the mosque behind rows of
police officers outside the building, waving two Swedish flags
while blasting the countrys national anthem through a speaker
system. He then started desecrating the Quran by tearing it up and
lighting it on fire with the cigarette he was holding.

In a recent community call, the team behind the Ethereum Name Service (ENS)
revealed plans to adopt a Layer-2 platform in light of increasing
gas fees on the mainnet. In response to this planned move, ENS
prices climbed roughly 10%, floating higher with increasing trading
volumes, evident in the daily chart.
The Ethereum Name Service
ENS is a decentralized naming system on Ethereum. It provides an
easy-to-use and human-readable naming system that allows users to
rename mainnet addresses, smart contracts, and other resources.
This system permits users to create and employ memorable domain
names instead of complicated addresses.
The team behind the ENS believes their solution makes it easier
for everyone to interact with the pioneer smart contracting
platform, and is handy, especially in sending and receiving
transactions. In this arrangement, the platform maps a users
domain, the human-readable address, with their original address or
deployed smart contract on the Ethereum mainnet.
However, since ENS maps all addresses on the Ethereum mainnet,
transaction fees apply, a concern considering the fluctuating gas
fees on Ethereum. In Ethereum, all transactions, including
transfers or smart contract deployment, must be paid for. The
amount paid depends on the level of demand in the network, and the
value tends to be volatile. Over the past few weeks, Ethereum gas
fees have been rising.
Fluctuating Gas Fees
According to YCharts data on June 30, the
average gas fee for sending a transaction stood at $34.62, down
from $42.77 recorded on June 5. As of May 12, it stood at $77.88.
The only time gas fees on Ethereum fell was on June 25, when it was
at $16.58. At this level, the gas fee is relatively high compared
to other networks. For example, the average transaction fee in
Bitcoin was $2.383 on June 30, down from $9.018 on May 12.
Meanwhile, in layer-2 platforms, transaction fees and smart
contract deployment costs are relatively lower, according to
trackers. As an illustration, gas fees on Optimism, a popular Ethereum
layer-2, is $0.03 as of July 2.
Considering the high gas fees on Ethereum, ENS, via two
improvement proposals, ENSIP-10 and EIP-3668, will further expand
to a Layer-2 platform as a relief for users sensitive to high gas
fees on Ethereum as part enhancing interoperability. During the

It appears that the co-founder of Ethereum, Vitalik Buterin,
doesnt trust the security of infrastructure allowing ETH staking.
Consequently, in a recent interview, Buterin stated that he would
only stake a limited amount of coins to ensure the network is
distributed and remain robust against malicious agents who might
try to take over the platform, reversing transactions.

Buterin has raised concerns about the potential risks of ETH
staking through third-party infrastructure, specifically regarding
the exposure of private keys and the danger it poses to his entire
stake. He believes that implementing a multi-signature system could
provide better protection. However, the current process is more
difficult to set up, leading to his increased caution.
In a multi-sig system, users have their private key to sign

Probably the biggest reason why I personally am not just staking
all of my ETH, that Im instead staking a fairly small portion, is
because if you stake your ETH, it has to be all out, like the keys
that access it have to be public on some system thats online, and
for safety, it has to be a multi-sig, and multi-sigs for staking
are still fairly difficult to set up, and it gets complicated in a
bunch of ways.

About 3 weeks ago my Wife-33 went into premature labor with our
daughter at 31 weeks. It was entirely unexpected and both my wife

My wife became preeclamptic among other serious complications
and for the first week we werent even sure if either my wife or our
daughter would make it. Luckily they both survived but our daughter
is still in the nicu after 3 weeks and probably will be in for

a very healthy and active person and even during most of her

When we think about rendering a portrait the typical utensils
come to mindpaint, pencils, and pens. However, using string and
nails, artists are able to mimic the depth achieved by line and
color. London-based artist Ben Koracevic has been practicing
string art
for years, sometimes spending hundreds of hours to achieve his
realistic illustrations. One of his newest projects merges string
art with sculpture in an innovative way.
The work in progress is inspired by the character Tommy (played
by Cillian Murphy) from the show Peaky Blinders. Koracevic
began the piece by forming the head with about 88 pounds (40
kilograms) of modeling clay wax and a few cans of expanding foam.
After the sculptural part was complete, he moved on to add 78,000
nails to the clay head. They were applied to every inch of the
surface at a relatively equal distance apart.
With the nails all in place, Koracevic was finally able to
incorporate his specialtyweaving the string. Just like his 2D
pieces, he deftly wraps the black string around the nails. This
creates a uniform color to the sculpture until Koracevic
crisscrossing thread over itself in key areas, ultimately resulting
in shadowy, opaque tones. While still in the works, the bust
already has an amazing amount of realism, like eyebrows, irises,
and pupils. Koracevic used 5,000 meters of string so far, but the
bust is not finished yet. Once it is complete, he intends to encase
the entire head in epoxy resin.

AITA? I didnt attend my sons wedding, I instead spent the
evening with his ex wife.
Quick backstory, after graduating high school my son moved 3
states away for college.
At 19 he married a girl he met, I tried convincing him to wait
because I personally felt he was too immature. They both dropped
out and moved back here to his hometown. At 20 they had their first
child, a beautiful little girl. 16 months later, my DIL gave birth
to their second child, a little boy.
After the first baby, my wife and I noticed our DIL wasnt happy.

visit, it was then she began unloading on us.
I know theres two sides to every story, but considering I know
my son, I believed her. I sat my son down numerous times to speak
with him regarding his marriage. He refused to take responsibility,
blamed her for everything even when I directly pointed out where he
was the sole problem.
They got into counseling, for a year things were ok on the
surface. Our DIL filed for divorce, my son 3 days later was on

that in an SHTF situation, a simple blister weve all had one could
actually be the death of you. Left untreated the risk of septicemia
rises exponentially.
The Personal First Aid Kit
This is the basic Boy Scout personal first aid kit and is
perfect for your everyday carry.
Two 4-by-4-inch sterile gauze pads
One small roll of adhesive tape
One 3-by-6-inch piece of moleskin
One small bar of soap or travel-size bottle of hand
sanitizer
One small tube of antibiotic ointment
One pair of scissors
One pair of non-latex disposable gloves
One CPR breathing barrier
Pencil and paper
6 band-aids
1 tube of Locktite Gel Super Glue
4 feet of duct tape (Wrapped around the pencil)
Why duct tape? Its perfect to remove those little bitty cactus
spines, makes a great pressure bandage, and a wrap for a sprained
ankle or splint.

If you are part of a small group, youll want a larger kit for

complaints and may even leverage misdiagnoses given by mental

psychiatric diagnosis. This leads to the patient being perceived as
untrustworthy and can protect the doctor legally no matter what
iatrogenic harms have been committed. These dishonest practices aid
in normalizing bad procedures and victim blaming.

Students at a Pakistani university received a disciplinary
notice for participating in Holi celebrations at their campus,
sparking debate on celebrating religious activities from other
faiths in the Muslim-majority nation.
Pakistan's Higher Education Commission bans
Holi celebrations across all educational institutes saying that
such activities portray a complete disconnect from the countrys
sociocultural values and are an erosion of the countrys Islamic
identity, reports Aaj News.
The Holi celebrations were performed at Quaid-i-Azam University
(QAU), a state-funded higher education institution (HEI) in the
countrys capital, Islamabad. A non-political cultural student
organization, Mehran Students Council, organized the event.
An official social media page posting news of the university
uploaded a video showing students dancing joyfully while
celebrating Holi, which is usually celebrated in the Indian
subcontinent every 12th month of the Hindu calendar, corresponding
to February or March in the Gregorian calendar.

OLYMPIA, Wash. Residents of Washingtons capitol city are
reporting an influx of tourism due to Rancids recently launched GPS
app which reroutes all users to the Pacific Northwest, frustrated
users confirmed.
I worked closely with the band to make sure their Avenue and
Alleyways GPS system found the best route to get to Olympia from
anywhere in the country, said the apps developed Mark Skwirm
Bartlett. Its like Waze or Google Maps for people who havent sold
out. The app has user input about which rest stops are easiest to
steal from and abandoned buildings you can squat in during your
trip. Also, the app always plays Rancid songs on a loop and you
cannot shut it off no matter how hard you try. The band insisted on
that feature.
Early users of the app admit they were frustrated by the fact
they keep ending up in Washington.
I was using the app to get to a wedding in Arizona where Im the
best man, said Kenny Kirkley. I zoned out a bit shortly after
hitting I-90 and before I realized it I was crossing into
Washington. The wedding is tomorrow and even if I drove through the
night I wouldnt make it in time, and if I use this fucking app it
would just make me do a circle and drop me right back in front of

I cant stand when bands churn out the same album
repeatedly, but I also hate when bands change their sound in any
way. I dislike Old Crows/Young Cardinals for both reasons. OC/YC
finds Alexisonfire retreading similar ground explored on Crisis but
with more of an alt-rock influence than ever before. Its not a
favorite of mine. Im not a lyrics guy so maybe this albums words
are amazing and they mean a lot to you, but Zeus gave me a brain
that doesnt understand lyrics after even 200 listens so sorry. This
could be a concept album about comparing various Cheez-It flavors
and it wouldnt affect my enjoyment in the slightest. (White Cheddar
is the best.)

Honorable Mention: Dogs Blood EP (2010)
This EP finds Alexisonfire rediscovering some youthful
recklessness after the relatively restrained and mature Old
Crows/Young Cardinals. Youll find 4 pretty solid songs here with a
lot of long guitar solos and instrumental passages. Possibly too
many. Also, this EP may have had a hand in popularizing that
colorful, cartoon-y art style that dominated post-hardcore and
metalcore for the next half-decade. Minus five points.

the Yazidi faith, prosecutors said when the trial started in

The Pakistani government entered into an agreement with a
radical Islamist party to try blasphemy suspects under the countrys
anti-terrorism laws along with existing anti-blasphemy laws in the
Muslim-majority nation.
Pakistani authorities made these concessions with the far-right
Tehreek-e-Labbaik Pakistan (TLP) after the party mounted a
25-day-long march protest at Sarai Alamgir in the countrys Gujrat
district, some 200 kilometers away from Lahore. The Islamist party
ended their protest after signing a pact with the ruling Pakistan
Muslim League (Nawaz) on June 17th.
Under the 12-point agreement they signed with the TLP, Pakistani
authorities agreed to arrest those accused of blasphemy and charge
them under the countrys Anti-Terrorism Act of 1997, alongside
Section 295-C of the Pakistani Penal Code, which prohibits
derogatory remarks against the Prophet Muhammad.

Two Filipino stand-up comedians are facing blowback from
religious believers after making jokes about their beliefs and
faith.
Gold Dagal and Jeleen Cubillas of the Comedy Manila group are
constantly being chastised online by members of the famed Iglesia
ni Cristo (or Church of Christ) and the Catholic Church,
respectively, after their stand-up comedy performance videos went
viral on Philippine social media.
During her stand-up performance at the Cine Adarna of the
University of the Philippines-Diliman, Cubillas described the
Catholic Church as the only religion with a "gang sign" referring
to the sign of the cross. Her performance video, uploaded on her
Facebook account on May 21st, garnered over 108,000 Angry and over
8,500 Sad reactions.
Dagal incorporated jokes about the Iglesia ni Cristo (also
called INC in the Philippines) during his stand-up performance,
describing the Church as "cool 'to" (English for It's cool or this
is cool and a play on the word kulto, which means cult in Tagalog)
because they have air-conditioned chapels.
He also made jokes about Eduardo Manalo, the current Executive
Minister of the Iglesia ni Cristo and grandson of Felix Manalo, the
churchs founder. Dagal joked about how easy it was for INC members
to sin because Eduardo looked like an unsuccessful lawyer,
pertaining to how he looks in the photos of him in a members

Polyphenol Epigallocatechin-3-Gallate on Markers of Inflammation

Inhalation of silica particles causes inflammatory changes leading
to fibrotizing silicosis. Considering a lack of effective therapy,
and a growing information on the wide actions of green tea
polyphenols, particularly epigallocatechin-3-gallate (EGCG), the
aim of this study was to evaluate the early effects of EGCG on
markers of inflammation and lung fibrosis in silicotic rats. The
silicosis model was induced by a single transoral intratracheal
instillation of silica (50 mg/mL/animal), while controls received
an equivalent volume of saline. The treatment with intraperitoneal
EGCG (20 mg/kg, or saline in controls) was initiated the next day
after silica instillation and was given twice a week. Animals were
euthanized 14 or 28 days after the treatment onset, and the total
and differential counts of leukocytes in the blood and
bronchoalveolar lavage fluid (BALF), wet/dry lung weight ratio, and
markers of inflammation, oxidative stress, and fibrosis in the lung
were determined. The presence of collagen and smooth muscle mass in
the walls of bronchioles and lung vessels was investigated
immunohistochemically. Early treatment with EGCG showed some
potential to alleviate inflammation, and a trend to decrease
oxidative stress-induced changes, including apoptosis, and a
prevention of fibrotic changes in the bronchioles and pulmonary
vessels. However, further investigations should be undertaken to
elucidate the effects of EGCG in the lung silicosis model in more
detail. In addition, because of insufficient data from EGCG
delivery in silicosis, the positive and eventual adverse effects of
this herbal compound should be carefully studied before any
preventive use or therapy with EGCG may be recommended.

acid (CGA) and epigallocatechin-3-gallate (EGCG) are major
polyphenolic constituents of coffee and green tea with beneficial
health properties. In this study, we evaluated the gut protecting
effect of CGA and EGCG, alone or in combination, on
D-galactose-induced aging mice. CGA plus EGCG more effectively
improved the cognition deficits and protected the gut barrier
function, compared with the agents alone. Specifically, CGA plus
EGCG prevented the D-galactose mediated reactive oxygen species
accumulation by increasing the total antioxidant capacity, reducing
the levels of malondialdehyde, and suppressing the activity of the
antioxidant enzymes superoxide dismutase and catalase. In addition,
supplementation of CGA and EGCG suppressed gut inflammation by
reducing the levels of the proinflammatory cytokines TNF, IFN,
IL-1and IL-6. Moreover, CGA and EGCG modulated the gut microbiome
altered by D-galactose. For instance, CGA plus EGCG restored the
Firmicutes/Bacteroidetes ratio of the aging mice to control levels.
Furthermore, CGA plus EGCG decreased the abundance of
Lactobacillaceae, Erysipelotrichaceae, and Deferribacteraceae,
while increased the abundance of Lachnospiraceae, Muribaculaceae,
and Rikenellaceae, at the family level. In conclusion, CGA in
combination with EGCG ameliorated the gut alterations induced by
aging, in part, through antioxidant and anti-inflammatory effects,
along with its gut microbiota modulatory capacity.

rats. Abstract: ETHNOPHARMACOLOGICAL RELEVANCE: The plant,
Ficus religiosa (L.) from the family Moraceae, has been extensively
used in Ayurveda and Unani. Traditionally this plant is known for
the treatment of constipation, liver diseases and neurological
disorders that are related to hypothyroidism.AIM OF THE STUDY: This
study was primarily designed to evaluate the effect of Ficus
religiosa leaf (FL) extract in ameliorating hypothyroidism in rats
and to identify the major bioactive compounds in the test extract
that might be responsible for the thyroid-altering activity. In
addition, the probable mechanism underlying the thyroid regulation
of the main FL constituents were analyzed by molecular
docking.MATERIALS AND METHODS: Adult female Wistar rats were used.
LC-ESI-MS/MS was performed to identify the compounds present in the
extract. HPLC analysis of FL extract was also performed. A pilot
study was made using 3 doses of FL extract. Out of 50, 100, and
200 mg/kg, 100 mg/kg appeared to be the most effective
one as it could increase thyroid hormones and decreased TSH levels.
In the final experiment, propyl-thiouracil (PTU)-induced
hypothyroid rats were orally treated with FL extract
(100 mg/kg) or L-thyroxine (100 g/kg, i.p.) daily for 28
consecutive days. On 29th day, all rats were sacrificed and the
serum levels of triiodothyronine (T), thyroxine (T), thyrotropin
(TSH), tumor necrosis factor-(TNF-), interleukin-6 (IL-6) and
hepatic 5' deiodinase-1(5'D1) were estimated by ELISA. Liver marker
enzymes (alanine aminotransferase, ALT and aspartate
aminotransferase, AST); total cholesterol (TC) and triglycerides
(TG); hepatic lipid peroxidation (LPO) and the activities of
antioxidants such as superoxide dismutase (SOD), catalase (CAT),
glutathione peroxidase (GPx), and glutathione (GSH) content were
estimated in liver tissues.RESULTS: LC-MS-MS analyses of the leaf
extract identified 11 compounds including the three major
compounds, betulinic acid (BA), chlorogenic acid (CGA), and quinic
acid (QA). While the PTU treatment decreased the levels of thyroid
hormones and 5'D1 activity, it increased the TSH, ALT, AST, TNF-,
IL-6, TC, and TG levels. Furthermore, hepatic LPO significantly
increased with a decrease in reduced GSH, SOD, CAT, and GPx.
However, FL treatment in PTU-induced animals nearly reversed these
adverse effects and improved liver function by decreasing ALT, AST,
hepatic LPO and increasing the levels of antioxidants. FL not only

isoflavonoid intakes are associated with lower incidence of type 2
diabetes. Abstract: Adequate evidence supports beneficial
effects of plant-derived phytochemicals against type 2 diabetes
(T2D). Among phytochemicals, dietary flavonoids is one of the
superb candidates. The whole studies are carried out in Western
populations, so it is needed to investigate the risk of T2D by
dietary flavonoid intakes in ethnic origins and other regions to
confirm these relations. This study was conducted to investigate
whether the daily consumption of total flavonoid and its subclasses
can affect the incidence of type 2 diabetes (T2D) in the Iranian
population.Eligible adults (=6547) were selected from among
participants of the Tehran lipid and glucose study with an average
follow-up of 3.0 years. Dietary intakes were assessed using a valid
and reliable 168-item semi-quantitative food frequency
questionnaire. Multivariate Cox proportional hazard regression
models were used to estimate the development of T2D in relation to
total intake of flavonoids.This study was conducted on 2882 men and
3665 women, aged 41.314.6 and 39.013.4 years, respectively. After
adjustment for several potential confounders (age, sex, diabetes
risk score, physical activity, energy, fiber and total fat
intakes), risk of T2D decreased from tertiles 1 to 3 for flavonols
(HR (95% CI): 1.00, 0.86 (0.64-1.16), 0.87 (0.63-0.93), P=0.01) and
isoflavonoids (HR (95% CI): 1.00, 0.84 (0.62-1.13), 0.64
(0.46-0.88), P=0.02), whereas non-significant results were found
for total flavonoid and other subclasses of flavonoid.These results
emphasize the potential protective role of flavonols and

is involved in Alzheimer's disease and other tauopathies. Recently,
tau has been shown to undergo liquid-liquid phase separation
(LLPS), which is implicated in the physiological function and
pathological aggregation of tau. In this report, we demonstrate
that the green tea polyphenol (-)-epigallocatechin-3-gallate (EGCG)
promotes the formation of liquid tau droplets at neutral pH by
creating a network of hydrophobic interactions and hydrogen bonds,
mainly with the proline-rich domain of tau. We further show that
EGCG oxidation, tau phosphorylation, and the chemical structure of
the polyphenol influence the efficacy of EGCG in facilitating tau
LLPS. Complementary to the inhibitory activity of EGCG in tau
fibrillization, our findings provide novel insights into the
biological activity of EGCG and offer new clues for future studies
on the molecular mechanism by which EGCG alleviates
neurodegenerative diseases.

Epigallocatechin-3-Gallate on Matrix Metalloproteinases in Terms of
Its Anticancer Activity. Abstract: Epidemiological studies
have shown that the consumption of green tea has beneficial effects
against cancer. Basic studies have provided evidence that
epigallocatechin gallate (EGCG) is a major contributor to these
effects. Matrix metalloproteinases (MMPs) are zinc-dependent
metalloproteinases with the ability to degrade the extracellular
matrix proteins and are involved in various diseases including
cancer in which MMPs have a critical role in invasion and
metastasis. In this review, we discuss the effects of EGCG on
several types of MMPs in the context of its anticancer activity. In
the promoter region, MMPs have binding sites for at least one
transcription factor of AP-1, Sp1, and NF-B, and EGCG can
downregulate these transcription factors through signaling pathways
mediated by reactive oxygen species. EGCG can also decrease nuclear
ERK, p38, heat shock protein-27 (Hsp27), and-catenin levels,
leading to suppression of MMPs' expression. Other mechanisms by
which EGCG inhibits MMPs include direct binding to MMPs to prevent
their activation and downregulation of NF-B to suppress the
production of inflammatory cytokines such as TNFand IL-1. Findings
from studies on EGCG presented here may be useful in the
development of more effective anti-MMP agents, which would give
beneficial effects on cancer and other diseases.
Still, if anything in that book was false, Kennedy would have
been sued to high heaven by now, Rogan reasoned. Reading that book
opened Rogans eyes to the fact that what were told by public
officials and the media isnt necessarily the truth. He also
realized just how easy it is to fall for a false narrative
especially when its all youre ever allowed to really hear.
For the first several minutes of the interview, Kennedy reviewed
his background and how he got to where he is today. He started his
legal career as an environmental lawyer in the mid-1960s, suing 500
polluters who had turned the Hudson River into a sewer, on behalf
of commercial fishermen whose livelihoods were threatened.
As a result of those lawsuits, the Hudson River was cleaned up
and restored. These successes led Kennedy to found
Riverkeeper,2 which
patrols waterways in 46 countries, holds polluters accountable and
defends clean drinking water.
How Kennedy Got Involved With Vaccine Safety
I would strongly encourage you to listen to this interview as
around the 10-minute mark Rogan asks Kennedy how he got into the
vaccine controversy. Although Kennedy had presented some of his
comments in the media previously, this is the first time he was
ever allowed to give his uninterrupted one-hour fascinating story
on major media.
In 2005, Kennedy started suing coal-burning powerplants and
cement kilns for releasing mercury into waterways. He launched
these lawsuits on behalf of local Riverkeeper chapters after
learning that mercury was being found in the flesh of most
freshwater fish. Pregnant women were also found to have levels that
might put their children at risk of developmental problems.
Kennedy also pushed legislation to protect children against
mercury and gave lectures on its dangers. During those lectures,
mothers started approaching him saying they suspected that mercury
(thimerosal) in the childhood vaccines had injured their children.
They told him that if he was really interested in protecting
children against mercury, he had to investigate vaccines.
Ted Kaczynski, better known as the Unabomber, earned notoriety
for committing 16 bombings between 1978 and 1995. But before he
turned violent, Kaczynski was used as a pawn by the Central
Intelligence Agency (CIA), which subjected him to cruel,
mind-altering experiments.1
Kaczynski, then, could be described as a construct of the CIA, a
product of its teachings. Three people lost their lives as a result
of Kaczynskis bombings, and 23 were injured, many seriously. He
died in his North Carolina prison cell in June 2023, where he is
said to have committed suicide.2
For some, his death puts to rest an era of terror that sowed
fear into Americans. But many questions remain. Kaczynskis homemade
bombs were mailed to those he believed to be destroying society
with technological advances,3 and the damage done to Kaczynskis psyche by
the CIA may never be fully known.
Unabomber Was Part of CIAs MK-Ultra Program
Kaczynski was just 16 years old and already a student at Harvard
University when he became part of the CIAs top-secret MK-Ultra
project. MK-Ultra involved mind control experiments, human torture
and other medical studies, including how much LSD it would take to
"shatter the mind and blast away consciousness."4 According to Kaczynskis
brother, David Kaczynski:5

Omega-3 The Healthy Fat Most People Don't Get Enough Of
One of the most important fats in the human diet is marine-based
omega-3. Unfortunately, in the past 100 years, our omega-6 intake
has nearly tripled, largely due to misleading or outright incorrect
marketing and government health campaigns, while our intake of
omega-3 has decreased 10fold, causing a severe imbalance in our
omega-3 to omega-6 ratio.
Ideally, you want to maintain a 4-to-1 ratio of omega-6 and
omega-3 fats or less, which is nearly impossible if you're
regularly eating processed foods or restaurant fare, as these are
loaded with omega-6 from industrial vegetable oils like corn oil
and canola oil. While you clearly need some omega-6s, they need to
be unprocessed (think whole, raw plant seeds and tree nuts), and
that cannot be said for most commercial vegetable oils.
One of the most significant dangers of vegetable oils is that
the damaged fats are integrated into your cell and mitochondrial
membranes, and once these membranes are impaired, it sets the stage
for all sorts of health problems.

BACKGROUND: Maternal diabetes compromises the quality and
developmental potential of oocytes. Therefore, it is important to
study how to ameliorate the adverse effects of diabetes on oocyte
quality. Epigallocatechin gallate (EGCG) has a variety of
physiological activities, including anti-inflammatory, antioxidant,
and anti-diabetes. In the present study, we evaluated the effect of
EGCG on the maturation of diabetic oocytes in vitro.OBJECTIVE:
Investigating the role of EGCG in restoring the adverse effects of
diabetes on oocyte quality.METHODS: Diabetes mouse model was
established by a single injection of streptozotocin (STZ). Oocytes
were collected and matured in vitro with/without EGCG in M16
medium.RESULTS: Compared with control, diabetic oocytes have a
higher frequency of spindle defects and chromosome misalignment,
but EGCG effectively reduces the incidence of oocytes with abnormal
spindle assembly and chromosome mismatches. Moreover, the abnormal
mitochondrial membrane potential (MMP) of diabetic oocytes is
significantly alleviated by EGCG, and the reduced expression of
genes regulating mitochondrial fusion (Mfn1 and Mfn2) and fission
(Drp1) in diabetic oocytes is significantly increased while EGCG is
added. EGCG also decreases the higher level of reactive oxygen
species (ROS) in diabetic oocytes that may be regulated by the
increased expression of superoxide dismutase 1 (Sod1) and
superoxide dismutase 2 (Sod2). EGCG can also reduce the DNA damage
of diabetic oocytes.CONCLUSIONS: Our results suggest that EGCG, at
least partially, improve the quality of diabetic oocytes.
Bitcoin had been trending over $31,000 for the better
part of Friday after reclaiming this support level from the bears.
However, this did not last long as the digital asset saw a flash
crash that sent its price spiraling toward the $29,000 level.
SEC Says ETF Filings Are Inadequate
On Friday, the Wall Street Journal released a
report that negatively impacted the market. The
report revealed that the United States Securities and Exchange
Commission thought that the recent barrage of Bitcoin ETH filings
that it had received was inadequate in their filings.
This follows a two-week stretch of positivity after
heavy hitters such as BlackRock, WisdomTree, and Fidelity, among
others, filed requests for Bitcoin Spot ETFs with the SEC.
Following the news of the filings, the price of Bitcoin rallied
tremendously, crossing $31,000 for the first time in over one
month.
However, with the news of the SEC saying that the
filings are inadequate, it has quickly eroded investor sentiment.
The price of BTC fell to $29,200 on Friday afternoon, before
recovering quickly. Although the damage was already done and BTC
bulls are now struggling to maintain support at $30,000.
Bitcoin In Freefall
Bitcoin had been able to establish a reasonably
bullish trend over the last couple of weeks. But that did not last
long as the SEC came to burst the bubble for investors. As a
result, Bitcoin seems to be in freefall with no reasonable support
to keep the price from falling lower.
In a matter of hours, the digital asset has lost
about $1,000 of its value, turning a previously good day into a
bloody day for investors. This rapid decline has shoved the
cryptocurrency below its 5-day moving average, which could see BTC
become bearish over the weekend.
However, in the mid and long-term, the digital asset
remains as bullish as ever, ranging above both its 50-day and
100-day moving averages. Both of these levels signal that while
bears may have gained temporary control of the price, bulls remain
largely in control. And as long as sell pressure remains low, then
BTC could recover back above $31,000 over the next couple of
days.
At the time of writing, BTC is down 1.59% in the last
24 hours to be trading slightly above $30,000. Although it is
currently struggling, its trading volume is sitting above $23.5
billion, showing a significant amount of demand for the digital
asset as well.

Abstract: Inflammation is pathogenic to skin diseases,
including atopic dermatitis (AD) and eczema. Treatment for AD
remains mostly symptomatic with newer but costly options, tainted
with adverse side effects. There is an unmet need for safe
therapeutic and preventative strategies for AD. Resveratrol (R) is
a natural compound known for its anti-inflammatory properties.
However, animal and human R studies have yielded contrasting
results. Mast cells (MCs) are innate immune skin-resident cells
that initiate the development of inflammation and progression to
overt disease. R's effects on MCs are also controversial. Using a
human-like mouse model of AD development consisting of a single
topical application of antigen ovalbumin (O) for 7 days, we
previously established that the activation of MCs by a bioactive
sphingolipid metabolite sphingosine-1-phosphate (S1P) initiated
substantial skin remodeling compared to controls. Here, we show
that daily R application normalized O-mediated epidermal
thickening, ameliorated cell infiltration, and inhibited skin MC
activation and chemokine expression. We unraveled R's multiple
mechanisms of action, including decreased activation of the
S1P-producing enzyme, sphingosine kinase 1 (SphK1), and of
transcription factors Signal Transducer and Activator of
Transcription 3 (Stat3) and NF-Bp65, involved in chemokine
production. Thus, R may be poised for protection against MC-driven
pathogenic skin inflammation.

loss and a subregion specific- reduction of serotonin reuptake
transporter induced by noise exposure in the central auditory
system. Abstract: Prolonged or excessive exposure to noise
can lead to hearing loss, tinnitus and hypersensitivity to sound.
The effects of noise exposure on main excitatory and inhibitory
neurotransmitter systems in auditory pathway have been extensively
investigated. However, little is known about aberrant changes in
neuromodulator systems caused by noise exposure. In the current
study, we exposed 2-month-old mice to a narrow band noise at 116 dB
SPL for 6 h or sham exposure, assessed auditory brainstem responses
as well as examined the expression of serotonin reuptake
transporter (SERT) in the cochlear nucleus (CN), inferior
colliculus (IC), and primary auditory cortex (Au1) using
immunohistochemistry. We found that noise exposure resulted in a
significant increase in hearing thresholds at 4, 8, 16, 24, and 32
kHz, as well as led to a significant reduction of SERT in dorsal
cochlear nucleus (DCN), dorsal IC (ICd), external IC (ICe), and Au1
layers I-IV. This reduction of SERT in these subregions of central
auditory system was partially recovered 15 or 30 days after noise
exposure. Furthermore, we examined efficacy of resveratrol (RSV) on
hearing loss and loss of SERT induced by noise exposure. The
results demonstrated that RSV treatment significantly attenuated
threshold shifts of auditory brainstem responses and loss of SERT
in DCN, ICd, ICe, and Au1 layers I-IV. These findings show that
noise exposure can cause hearing loss and subregion-specific loss
of SERT in the central auditory system, and RSV treatment could
attenuate noise exposure-induced hearing loss and loss of SERT in
central auditory system.

NAFLDexploring the hepatic NF-B/NLRP3 inflammasome pathway and
intestinal microbiota composition in high-fat and high-fructose

randomly assigned into 5 groups: Normal control group (NC); HFFD
group; HFFD with low dose of lycopene group (LLY, 20mg/kg/d); HFFD

against postmenopausal atherosclerosis progression through reducing
PCSK9 expression via the regulation of the ER-mediated signaling
pathway. Abstract: Elevated circulating proprotein convertase
subtilisin/kexin 9 (PCSK9) levels are an important contributor to
postmenopausal atherosclerosis (AS). We have previously reported
that resveratrol (RSV), as a phytoestrogen, reduces hepatocyte
steatosis and PCSK9 expression in L02 cells. This study aimed to
investigate how RSV reduces PCSK9 expression to inhibit
postmenopausal AS progression. Here, we found that treatment of
Ovx/ApoE -/- mice with RSV significantly reduced dyslipidemia,
plasma PCSK9 concentration and aortic plaque area. In addition, RSV
significantly inhibited liver fat accumulation and improved the
hepatocyte ultrastructure. Further studies showed that RSV
upregulated estrogen receptor(ER) expression, while reduced the
liver X receptor(LXR) expression and sterol
regulatory-element-binding protein-1c (SREBP-1c) transcriptional
activity. In vitro, RSV inhibited insulin-induced elevated
intracellular/extracellular PCSK9 levels, enhanced
receptor-mediated uptake of low-density lipoproteins in HepG2
cells. Furthermore, RSV attenuated the activity of the
SRE-dependent PCSK9 promoter. However, these effects can be
partially reversed by the antiestrogen ICI 182,780. Attenuation of
these changes with ERinhibition suggest that RSV may prevent the
progression of postmenopausal AS by reducing PCSK9 expression in
hepatocytes through ER-mediated signaling.

against sepsis-associated encephalopathy by inhibiting
NLRP3/Caspase-1/GSDMD pyroptosis pathway and reducing blood-brain
barrier damage. Abstract: Puerarin (Pue), an isoflavone
compound extracted from Pueraria, has been shown to inhibit
inflammation and reduce cerebral edema. The neuroprotective effect
of puerarin has attracted much attention in recent years.
Sepsis-associated encephalopathy (SAE) is a serious complication of
sepsis that causes damage to the nervous system. This study aimed
to investigate the effect of puerarin on SAE and elucidate the
potential underlying mechanisms. A rat model of SAE was established
by cecal ligation and puncture, and puerarin was injected
intraperitoneally immediately after the operation. Puerarin was
found to improve the survival rate and neurobehavioral score of SAE
rats, alleviate symptoms, inhibit the level of brain injury markers
NSE and S100, and improve the pathological changes in rat brain
tissue. Puerarin was also found to inhibit the level of factors
related to the classical pathway of pyroptosis, such as NLRP3,
Caspase-1, GSDMD, ASC, IL-1, and IL-18. Puerarin also reduced the
brain water content and penetration of Evan's Blue dye in SAE rats,
and reduced the expression of MMP-9. In the in vitro experiments,
we further confirmed the inhibitory effect of puerarin on neuronal
pyroptosis by establishing a pyroptosis model in HT22 cells.
Our findings suggest that puerarin may improve SAE by inhibiting
the classical pathway of NLRP3/Caspase-1/GSDMD-mediated pyroptosis
and reducing blood-brain barrier damage, thus playing a role in
brain protection. Our study may provide a novel therapeutic
strategy for SAE.

Glycolysis and Increases Cisplatin Chemosensitivity in Oral
Squamous Cell CarcinomaFBXW7/mTOR Signaling. Abstract: This
study aimed to observe the effects of puerarin on glycolysis and
cisplatin sensitivity in oral squamous cell carcinoma (oSCC) cells
and to explore the underlying mechanisms. CAL27 cells over- or
under-expressing FBXW7 were treated with cisplatin or puerarin, and
the levels of proteins involved in glycolysis as well as the
activity of the respective enzymes were assessed. Glucose uptake
and lactate production were also evaluated, and the IC50 value of
cisplatin in CAL27 cells was determined. FBXW7 overexpression
significantly downregulated HK2, PKM2, and LDH; suppressed the
activity of the corresponding enzymes hexokinase, pyruvate kinase,
and lactate dehydrogenase; as well as reduced glucose uptake and
lactate production. FBXW7 overexpression was also associated with
decreased mTOR phosphorylation and increased cisplatin sensitivity.
These effects were partially antagonized by lactate or the mTOR
agonist MHY1485. Puerarin suppressed glycolysis by reducing glucose
uptake and lactate production, while it promoted cisplatin
sensitivity and activated the FBXW7/mTOR signal pathway in a
concentration-dependent manner. These effects were antagonized by
FBXW7 downregulation or treatment with MHY1485. Our results suggest
that FBXW7 improves cisplatin chemosensitivity and suppresses
glycolysis in oSCC cells, indicating its promising potential as a
target for puerarin to regulate the cisplatin sensitivity of oSCC
cells.

(Fer)-1 groups. Parameters related to ferroptosis included levels
of malondialdehyde (MDA), 4-hydroxynonenal (4-HNE), ATP, reactive
oxygen species (ROS), glutathione (GSH), prostaglandin endoperoxide
synthase (Ptgs) 2 mRNA, glutathione peroxidase (GPX) 4 protein and
iron. In H9C2 cells, puerarin or Fer-1 pretreatment reduced
ferroptosis, as indicated by decreased ROS and increased GSH, ATP
levels. In vivo, wild-type mice were randomly divided into sham,
I/R+vehicle, I/R+puerarin, and IR+Fer-1 groups. The I/R model was
established by 30min of left anterior descending artery occlusion
followed by 24h of reperfusion. Pretreatment with puerarin or Fer-1
significantly reduced infarct size in I/R mice, and decreased the
activities of Myeloperoxidase (MPO) and cardiac enzymes such as
creatine kinase MB isoenzyme (CK-MB), aspartate aminotransferase
(AST), and lactate dehydrogenase (LDH) compared to those in the
vehicle-treated group. Puerarin also reduced the production of MDA
and 4-HNE, reduced the mRNA expression of Ptgs2 mRNA, and increased
GPX4 protein expression. These results showed that puerarin exerted
protective effects against myocardial I/R injury by inhibiting
ferroptosis and inflammation, and therefore may have therapeutic
potential for treatment of acute myocardial infarction.

resveratrol nano-encapsulated supplements against breast cancer via
the regulation of oxidative stress. Abstract: AIM: Several
shreds of evidence have supported the growth-inhibitory effect of
resveratrol in breast cancer. Owing to the low efficiency, we aimed
to fabricate an ACN nanoparticle containing resveratrol to target
the proliferation of breast cancer cells.METHODS: Resveratrol
encapsulation was characterized using spectrophotometry, FTIR, and
SEM. The cytotoxicity and antioxidant activities of compounds were
evaluated by MTT, NO, FRAP, and qRT-PCR assays on MCF7 and SKBr3
cells.RESULTS: Our result found that the encapsulation efficiency
was 87%, the particle size was 20015nm, and the zeta potential was
310.4mV. The prepared RES+ACN had controlled in vitro release.
RES+ACN nanoparticle showed a significantly increased cytotoxicity
in both cell lines. The reduced level of NO and enhance
antioxidative content in both cells, especially the MCF7, were in
line with increased expression of Nrf2 and SOD as well as more
apoptotic effect.CONCLUSION: Decreased growth and increased
expression of Nrf2 in MCF7 compared to SKBr3 cells proved that the
upregulation of Nrf2 by nanoresveratrol is likely contributed to
its relationship with ER/PR signaling factors, albeit its precise
mechanism is needed to be more elucidated.

meta-analysis of 47 randomized controlled trials. Abstract:
OBJECTIVE: To evaluate safety and efficacy of dietary polyphenols
in the treatment of rheumatoid arthritis (RA).METHODS: CNKI,
Pubmed, Cochrane library, Embase were searched to collect
randomized controlled trials (RCTs) of dietary polyphenols in the
treatment of RA. The databases were searched from the time of their
establishment to November 8nd, 2022. After 2 reviewers
independently screened the literature, extracted data, and assessed
the risk of bias of the included studies, Meta-analysis was
performed using RevMan5.4 software.RESULTS: A total of 49 records
(47 RCTs) were finally included, involving 3852 participants and 15
types of dietary polyphenols (Cinnamon extract, Cranberry extract,
Crocus sativus L. extract, Curcumin, Garlic extract, Ginger
extract, Hesperidin, Olive oil, Pomegranate extract, Puerarin,
Quercetin, Resveratrol, Sesamin, Tea polyphenols, Total glucosides
of paeony). Pomegranate extract, Resveratrol, Garlic extract,
Puerarin, Hesperidin, Ginger extract, Cinnamon extract, Sesamin
only involve in 1 RCT. Cranberry extract, Crocus sativus L.
extract, Olive oil, Quercetin, Tea polyphenols involve in 2 RCTs.
Total glucosides of paeony and Curcumin involve in more than 3
RCTs. These RCTs showed that these dietary polyphenols could
improve disease activity score for 28 joints (DAS28), inflammation
levels or oxidative stress levels in RA. The addition of dietary
polyphenols did not increase adverse events.CONCLUSION: Dietary
polyphenols may improve DAS28, reduce C-reactive protein (CRP) and
erythrocyte sedimentation rate (ESR), and improve oxidative stress,
etc. However, more RCTs are needed to verify or modify the efficacy

PMID: 37049468 Abstract Title: Sesamin Metabolites Suppress
the Induction of Cellular Senescence. Abstract: Cellular
senescence induces inflammation and is now considered one of the
causes of organismal aging. Accumulating evidence indicates that
age-related deterioration of mitochondrial function leads to an
increase in reactive oxygen species (ROS) and DNA damage, which in
turn causes cellular senescence. Thus, it is important to maintain
mitochondrial function and suppress oxidative stress in order to
inhibit the accumulation of senescent cells. Sesamin and its isomer
episesamin are types of lignans found in sesame oil, and after
being metabolized in the liver, their metabolites have been
reported to exhibit antioxidant properties. However, their effects
on cellular senescence remain unknown. In this study, the effects
of sesamin, episesamin, and their metabolites SC1 and EC1-2 on
replicative senescence were evaluated using human diploid lung
fibroblasts, and TIG-3 cells. The results showed that sesamin and
episesamin treatment had no effect on proliferative capacity
compared to the untreated late passage group, whereas SC1 and EC1-2
treatment improved proliferative capacity and mitigated DNA damage
of TIG-3 cells. Furthermore, other cellular senescence markers,
such as senescence-associated secretory phenotype (SASP),
mitochondria-derived ROS, and mitochondrial function (ROS/ATP
ratio) were also reduced by SC1 and EC1-2 treatment. These results
suggest that SC1 and EC1-2 can maintain proper mitochondrial
function and suppress the induction of cellular senescence.

and mechanisms of total glucosides of Paeony on Graves disease in
BALB/c mice]. Abstract: To investigate the protective effect
and its immunoregulatory mechanism of Total Glucosides of Paeony
(TGP) against Graves' Disease (GD) model on BALB/c mice.Fifty
female (6 weeks old, weighing 16-18 g) BALB/c mice of specific
pathogen free were divided into control group according to random
number table method, model group, early low-dose TGP intervention
group (250 mgkgd), early high-dose TGP intervention group (500
mgkgd), and late TGP intervention group, with 10 mice in each
group. Except the control group, the other 4 groups were immunized
3 times (0, 3, and 6week) with recombinant adenovirus expressing
the thyroid stimulating hormone receptor (TSHR) A subunit to
establish the GD model. The early low-dose and high-dose
intervention group were given diets containing different doses of
TGP throughout the whole process, and the late intervention group
was given diets containing low doses of TGP from the 1week after
the 2immunization (week 4). The levels of thyrotropin receptor
antibody (TRAb) and total thyroxine (TT4) were detected in the tail
venous blood of mice at the 4week. At the 10week, the serum TRAb
and TT4 levels and the ratio of regulatory T cells (Treg) in each
group were detected, and the pathological changes of thyroid tissue
were observed. Serum helper T cell 1(Th1) and Th2 cell-related
factors interleukin-2 (IL-2), IL-4, IL-5, IL-10, IL-12p70,
granulocyte-macrophage colony-stimulating factor (GM-CSF),
interferon-(IFN-) and tumor necrosis factors-(TNF-) were detected
to investigate the protective effect of TGP on GD model in BALB/c
mice and its mechanism.At the 4week, The level of TT4
[(55.0712.89)g/L] in early high-dose intervention group was lower
than that in model group [(74.338.63)g/L] (all0.05). TRAb level of
mice between early low-dose, early high-dose, late intervention
groups and model group was no significant difference (all>0.05).

hepatoprotective activity of total glycosides of paeony with high
content of paeoniflorin extracted from Paeonia lactiflora Pall.
Abstract: The aims of this work were to obtain total
glucosides of paeony (TGP) with high content of paeoniflorin and
evaluate the hepo-protective properties of TGP. After optimization
by response surface methodology, optimized conditions were as
follows: extraction time 33.0 min, extraction temperature
48 C, ethanol content 44%, and the yield of TGP was
47.68 mg/g. Moreover, under established macroporous resin
purification, paeoniflorin content of TGP achieved 67.6% in
1.5 L scale-up verification experiment. Purified TGP (p-TGP)
was further analyzed by UHPLC-Q-Orbitrap-MS/MS, and 35 compouds
including paeoniflorin were identified. The obtained p-TGP
effectively reduced biochemical indexes and inflammatory cytokines
in liver tissue of acute alcoholic liver injury mice model.
Depending on this work, TGP with definitive compound composition
exhibited great protective effect against acute alcoholic liver
injury in vivo. Furthermore, the finding of this work will be
helpful to understand the relationship between compound composition
and functional properties of Chinese herb extracts.

Paeoniflorin is one of the important components inplants. In this
study, we usedas a model host andas a bacterial pathogen to
investigate the possible role of paeoniflorin treatment
againstinfection in the host and the underlying mechanisms.
Posttreatment with 1.25-10 mg/L paeoniflorin could
significantly increase the lifespan ofinfected nematodes. After the
infection, thecolony-forming unit (CFU) andaccumulation in
intestinal lumen were also obviously reduced by 1.25-10 mg/L
paeoniflorin treatment. The beneficial effects of paeoniflorin
treatment in increasing lifespan ininfected nematodes and in
reducingaccumulation in intestinal lumen could be inhibited by RNAi
of,, and. In addition, paeoniflorin treatment suppressed the
inhibition in expressions of,, andcaused byinfection in nematodes,
suggesting that paeoniflorin could increase lifespan ofinfected
nematode by activating PMK-1, EGL-1, and BAR-1. Moreover, although

2.5-10 mg/L paeoniflorin reduced the cyclic-di-GMP (c-di-GMP)
level. Therefore, paeoniflorin treatment has the potential to
extend lifespan ofinfected hosts by reducing bacterial accumulation

against cisplatin-induced acute kidney injury through targeting
Hsp90AA1-Akt protein-protein interaction. Abstract:
ETHNOPHARMACOLOGICAL RELEVANCE: Paeonia lactiflora Pall has been
used in Chinese Medicine for thousands of years, especially having
anti-inflammatory, sedative, analgesic and other ethnic
pharmacological effects. Moreover, Paeoniflorin is the main active
ingredient of the Paeonia lactiflora Pall, and most are used in the
treatment of inflammation-related autoimmune diseases. In recent
years, studies have found that Paeoniflorin has a therapeutic
effect on a variety of kidney diseases.AIM OF THE STUDY: Cisplatin
(CIS) is limited in clinical use due to its serious side effects,
such as renal toxicity, and there is no effective method for
prevention. Paeoniflorin (Pae) is a natural polyphenol which has a
protective effect against many kidney diseases. Therefore, our
study is to explore the effect of Pae on CIS-induced AKI and the
specific mechanism.MATERIALS AND METHODS: Firstly, CIS induced
acute renal injury model was constructed in vivo and in vitro, and
Pae was continuously injected intraperitoneally three days in
advance, and then Cr, BUN and renal tissue PAS staining were
detected to comprehensively evaluate the protective effect of Pae
on CIS-induced AKI. We then combined Network Pharmacology with
RNA-seq to investigate potential targets and signaling pathways.
Finally, affinity between Pae and core targets was detected by
molecular docking, CESTA and SPR, and related indicators were
detected in vitro and in vivo.RESULTS: In this study, we first
found that Pae significantly alleviated CIS-AKI in vivo and in
vitro. Through network pharmacological analysis, molecular docking,
CESTA and SPR experiments, we found that the target of Pae was Heat
Shock Protein 90 Alpha Family Class A Member 1 (Hsp90AA1) which
performs a crucial function in the stability of many client
proteins including Akt. RNA-seq found that the KEGG enriched
pathway was PI3K-Akt pathway with the most associated with the
protective effect of Pae which is consistent with Network
Pharmacology. GO analysis showed that the main biological processes
of Pae against CIS-AKI include cellular regulation of inflammation
and apoptosis. Immunoprecipitation further showed that pretreatment
with Pae promoted the Hsp90AA1-Akt protein-protein Interactions
(PPIs). Thereby, Pae accelerates the Hsp90AA1-Akt complex formation
and leads to a significant activate in Akt, which in turn reduces
apoptosis and inammation. In addition, when Hsp90AA1 was knocked
down, the protective effect of Pae did not continue.CONCLUSION: In

glucosides of paeony in the treatment of Sjogren syndrome: A
network pharmacology study. Abstract: BACKGROUND: We aimed to
explore the underlying mechanism of the total glucoside of peony
(TGP) in treating Sjogren syndrome (SS) using the network
pharmacology approach.METHODS: The protein targets of TGP and SS
were identified by database search. Then, the intersection of the
two groups was studied. The drug-target network between TGP and the
overlapping genes was constructed, visualized, and analyzed by
Cytoscape software. Gene Ontology and Kyoto Encyclopedia of Genes
and Genomes enrichment were performed to analyze these genes.
Finally, the predictions of potential targets were evaluated by
docking study.RESULTS: Forty-six overlapping genes were discovered.
The results suggested that TGP used in the treatment of SS is
associated with cellular tumor antigen p53, neurotrophic tyrosine
kinase receptor type 1, and epidermal growth factor receptor, as
well as their related 3372 protein networks, which regulate
intrinsic apoptotic signaling pathway, cellular response to
oxidative stress, rhythmic process, and other processes. Molecular
docking analysis proved that hydrogen bonding is the main form of
interaction.CONCLUSION: Our research provided the protein targets
affected by TGP in SS treatment. The key targets (caspase 3,
vascular endothelial growth factor A, glyceraldehyde-3-phosphate
dehydrogenase, etc.), which involve 3372 proteins, are the
multitarget mechanism of TGP in SS treatment.

I cant stand when bands churn out the same album
repeatedly, but I also hate when bands change their sound in any
way. I dislike Old Crows/Young Cardinals for both reasons. OC/YC
finds Alexisonfire retreading similar ground explored on Crisis but
with more of an alt-rock influence than ever before. Its not a
favorite of mine. Im not a lyrics guy so maybe this albums words
are amazing and they mean a lot to you, but Zeus gave me a brain
that doesnt understand lyrics after even 200 listens so sorry. This
could be a concept album about comparing various Cheez-It flavors
and it wouldnt affect my enjoyment in the slightest. (White Cheddar
