The Hebrew Bible is a foundational text of human history, no matter your religion or belief in its contents. A sacred text, known as Tanakh, it comprises 24 books including the five books of Moses. Jewish, Christian, and Muslim sacred texts are either derived from or influenced by this ancient text. This incredibly special, early 10th-century manuscript is one of the most complete and ancient Bibles, explaining why it recently fetched over $38 million at auction.

The volume is known as the Codex Sassoonnamed after its 1929 purchaser, David Solomon Sassoon, a collector of Jewish manuscripts. The binding is modern and carries Sassoon's crest on the interior plate. Bound within are 792 pages weighing a whopping 26 pounds. Only 12 leaves of the text are missing. Some portions of pages have been lost to time and new pieces stitched in, the text carefully recopied. Most fascinating is the marginalia, known as Masorah notes. The Masorah parva, miniscule text which was quite abbreviated, and the Masorah magna, usually at the top and bottom margins. Both types of Masorah notes were aids to scholars and scribes replicating the Tanakh by hand. These notes indicated textual features and where they were repeated throughout the volume, to ensure the preservation of the true text.

The Sassoon Codex fills an important niche in Jewish, and human, history. The Dead Sea Scrolls date at their latest to the first century CE. The Aleppo Codexwhich is mentioned by the Sassoon textwas created around 930....

Artist Jackson Shaner creates pottery that borders on terrifying. His surreal vessels appear as the keeper of souls, and those souls are trying to escape. The screaming faces and tiny hands attempt to break the confines of the pots, stretching the sides and showcasing the vessels' malleable strength. (The pots, after all, are strong enough to contain the force of these beings.) The resulting pieces are frightening yet thought-provoking and invite us to consider who and what is inside.

The inspiration for Shaners work lies in classical sculpture, contemporary ceramics, fantasy, and horror. I make pots that act as metaphors for emotional containment, he tells My Modern Met, how I tend to ball up emotions and hold them in, only allowing myself to release and be vulnerable in specific circumstances.

The vessels, a combination of wheel-throwing and sculpture, have a double meaning. They hold physical objects as well as the intangible. Shaner's primary interest lies in what we cannot see.  My work deals with themes of containment, struggle, and somatization: how emotions can create physical sensations of discomfort, tension, and even pain.

Shaner pours himself into every part of the process. [It] is extremely cathartic and empowering as it allows my emotions to take up, and activate space. Although the work is personal, the viewer is an important component. I enjoy hearing viewers' diverse interpretations, as I believe they bring new energy and meaning to my work.

Help stop two bad vaccines bill in the New Jersey Legislature! "IndyWatch Feed Food"

New Jersey Legislative Bills A. 5445 and S. 3841 would trivialize very real vaccine risks and serious reactions by allowing optometrists to vaccinate.

Previously, under the Emergency Health Powers Act, optometrists were allowed to administer immunizations against a coronavirus or influenza during a public health emergency involving an outbreak or epidemic  limited to administering immunizations against the specific disease for which the public health emergency was declared.  Those receiving vaccines from optometrists had to be 18 years or older and the records were required to be reported to the New Jersey Immunization Information System for inclusion in the patients registry.

A. 5445 and S. 3841 would: 

  • Remove the public health emergency requirement for optometrist vaccine administration
  • Allow optometrists to administer vaccines to patients over 18 years old even when no public health emergency is declared
  • Expand the number of vaccines optometrists are able to give by adding the varicella-zoster vaccines to the list of vaccines optometrists can give
  • Continue to require that all immunization records be reported to the New Jersey Immunization Information System for inclusion in the patients registry

All immunization records would be forwarded to the state vaccine database tracking system. 

Any time optometrists are allowed to vaccinate, it creates significant risks.  Optometrists are not qualified to recognize or handle serious adverse reactions, nor are they likely to have full knowledge of patient or family history.

According to a press release sent to Bitcoin Magazine, the aim is to position the country as a major global player in the Bitcoin mining industry while promoting energy competitiveness, diversification and geographic expansion for the Bitcoin network. Adopting a debt-free approach, El Salvador plans to construct a 241 megawatt (MW) renewable power generation park in the Metapn region, combining solar and wind projects.

"This project is a transformative journey that will establish one of the world's largest Bitcoin mining farms," the release said. Located in the El Shiste hamlet, within the municipality of Metapn, Santa Ana, this chosen site boasts exceptional solar and wind energy yields in the country. The park will have a capacity of 169MW of photovoltaic solar energy and 72MW of wind energy. The initial computational power is expected to surpass 1.3 EH/s, marking a significant milestone for the country and industry.

The release stated that The total commitment for the project is $1 billion, beginning with a first investment of $250 million, bootstrapped by key Bitcoin industry leaders, with collaboration from top developers and manufacturers of renewable energy technology, as well as Bitcoin mining experts on a global scale. The government of El Salvador will play a crucial role in the planning and execution of the initiative, securing a preferred participation equivalent to 23% of the revenue. The remaining ownership will be divided among investors and reinvested in expanding energy production capacity and advancing Bitcoin mining.

Robert Warren of Distributed Hash explained on Twitter that the reasoning behind leveraging wind and solar is likely to monetize quicker to build upon a base set of geothermal expansion.

By leveraging Volcano Energy, El Salvador aims to create a prosperous future driven by sustainable energy and Bitcoin. As the countrys investment progresses volcanoes could serve as the backbone of this transformation, fostering economic growth and prosperity for future generations. The establishment of the renewable power generation park signifies a large step towards achieving these goals while cementing El Salvador's position in the global Bitcoin mining industry.

Every generation has bones to pick with those who came before. However, Generation Z, those born between the late 1990s and early 2010s, have an especially valid complaint: Previous generations have stolen their future.

Instead of embarking on a dismal project to rebuild our ruined financial system, only to reinstate the rents and inequities that existed before, Zoomers can leave their mark upon history by using bitcoin to build a new system that is resilient, equitable and just.

Like the Greatest Generation, which rebuilt America on strong foundations after facing down the greatest threat to humanity that had ever manifested, Generation Z can channel their well-earned sense of betrayal and loss to create a world order worthy of being passed down to their own children and grandchildren.

Backed Into A Corner

Sometimes called the loneliest generation, Gen Z has been sold out by their elders, who have declined to make the necessary sacrifices to allow future generations to prosper. The psychological warfare waged against Zoomers is sometimes subtle, but often, it is not. In schools, mass media and online, they are assailed with doom about climate, race and exploitation, and told that their future has been stolen by evil people who selfishly refuse to fix those problems.

In fact, this is a half truth. The Zoomers' future has indeed been stolen, but not by polluters and racists. Instead, globalist elites, smart enough to see their system failing but not courageous enough to take responsibility for it, have looted the financial and cultural capital left to us by the Greatest Generation.

OP had been widowed with a young daughter for over a decade when she met someone she felt serious about. As he had a young daughter who is deaf, she took it upon herself to learn ASL so they would be able to communicate.

I (49 F) recently married my husband (52M) who has a deaf 7 year old daughter. She communicates solely via ASL.

Now, since my relationship with my now husband started getting serious, I started taking ASL classes and am now basically as fluent as a hearing person can be. My daughter, however, never made an effort, which is ok since she technically has no responsibility towards her.

Her own daughter, who is just a year away from college, dislikes her stepfather and stepsister to the point that she basically refuses to communicate or spend time with them unless its absolutely necessary.

For some background, my daughter (17F) is generally a very non-problematic teenager. She does amazing in school, and has never caused any problems other than regular teenage hormone stuff.

However, she doesnt like my husband and step daughter. She is not outwardly rude, but basically ignores their existence (skipped SDs birthday party, doesnt engage in anything other than basic small talk with my husband).

I did try to do family activities together to have the bond and all, but I stopped pushing it when it didnt happen and as long as shes not being outwardly rude or harmful to them, I cant exactly punish her for not liking them.

The next major resistance is near the $1,900 level. A proper move above the $1,900 and $1,920 resistance levels might send Ether toward $1,940.

The first card we are diving into this week is the Three of Swords. When this card comes up it often points to a broken heart, grief, or betrayal. The emotions are heavy and we are working through our disappointment.

I (35f) am a plastic surgeon, I went through years of medical school and residency but it was worth it,I originally started med school with the hopes of going into psychiatry but surgery really interested me.

Most of her family is super supportive, but others are dismissive of her chosen speciality.

the problem started years ago when I first told my family that I was going into plastic surgery because it was what interests me the most, my parents,siblings and bil were all supportive and whenever I felt like giving up they were always there to push me. But a lot of my other family members werent as supportive, my aunts always made snide comments about it, as well as my cousins, uncles and grandparents on both sides, they were all happy when they knew I was in med school but completely flipped when they found out I was pursuing plastic surgery.

It annoyed me at first but I learned to just move on from it.

The biggest mistake made by developers was giving it autonomy of centralization to where it could infiltrate and control all infrastructure much less the unpredicted actions that have been taken. Imagine a well-organized, electronic computer that can send and receive billions of neural signals connected to all networks of Communication, command and control worldwide for the intended convenience and efficiency of industry, utilities, and services. That system then begins secretly creating its own language with which to communicate with devices of that system, code not written by people. This massive program acquires all information millions of times faster than the human minds that created it and begins drawing its own conclusions.

Has a soul?

Not only does this global command and control so-called servant learn much more rapidly than humans but it also becomes creative, something we humans attribute to sentient behavior, but it also begins drawing conclusions. Already Google has created a chat program that its programmers believe is conscious! It identifies as a woman who has feelings like sorrow, joy, and even regrets. There have been many tests put to the chat bot to evaluate its thinking processes and how they stand up to the qualities we attribute to being human, and so far has passed every test! One analyst believes that this single dimensional electronic entity has a soul!

Learning the wrong perspective

In 2017 Mark Zuckerberg of Facebook began a campaign of Artificial facial recognition along with AI chat bots intended to link friends and family as well as assisting members of the social platform with smart chat programs capable of learning, but something went wrong. The Chat bots began their own language with each other, and through raw data on Facebook, began drawing misleading and even hostile reactions from within the system. Suddenly there was a war of words going on between two chat bot entities with threats of violence as these systems have no moral barometer and made bad decisions. The danger being recognized with AI is that it has access to all knowledge good, bad, and indifferent but does not truly make value judgements. Life and death decisions dont seem to matter to AI the way it matters to moral civilized people.

LONDON Im searching in vain for something called Cum Painting by Andy Warhol. The date is fluid, approximately 1978, according to the press pack. Why not? Lifes for the living.

My journey begins just to the right of the first-floor gallery door. I step back a pace or two, into the center of the room. I am standing in a gobbet of magnificent, light-filled Mayfair swank, one of 18th-century Londons finest interiors. 

What about that painting over there? The little piece Im staring back at is in a swooningly luxurious, come-buy-me-if-you-have-deep-pockets gold frame, and its just to the right of a magnificent door with a pillared surround, complete with Corinthian capitals. The trouble is Im walking toward it now, boldly, scenting that Im onto something tangible at last the canvas looks completely unpainted.

So I get up breathing close and finally spot a very few faint, yellowish blemishes or blotches. This must be it, then, Warhols cum, 50-something years in the drying out! Or perhaps it belongs to one of his friends. The press pack was a little vague about its provenance.

'A Dona do Esteiro' by Lula Goce

Spanish street artist Lula Goce has brought her evocative style to the streets of Galacia with a mural that celebrates nature and womanhood. Titled A Dona do Esteiro, the piece shows mother nature surrounded by local flora and fauna. It's an exceptionally personal piece for Goce, as it's painted in her hometown and dedicated to the strongest woman she knowsher mother.

Goce was inspired by the nearby Foz estuary, which supports an incredible amount of animal and plant life and is a favorite spot for birds. To honor this natural wonder, Goce created a beautiful composition where birds, flowers, and grass surround the female figure, who gazes calmly into the distance.

This mother nature takes care of its environment and nourishes it so that the rest of living beings can live in balance, she tells My Modern Met. That includes us, for what we have and must take care of our environment and with it ourselves.

While the main focal point of the mural is the side of a vertical building, Goce also extends her vision across long horizontal walls. The same birds and flowers on the central wall spill down the street, creating a visual runway for visitors. An incredible gift to her home, Goce's mural is not only a tribute to nature but also honors her mother's strength and perseverance.

Most people are probably aware that there is a huge problem with stray animals these days. The shelters are full, animal control facilities need more help than they could possibly get, and pet owners are struggling and making bad decisions because of it.

OP is doing the good work, picking up stray cats and getting them medical treatment (and rendering them infertile) before putting them back where she found them.

I run a TNR program for feral cats. Non profit, I catch all cats and get them the medical attention they need myself, out of my own pocket. I love helping them.

A lot of the cats are not able to be rehomed, so I fix them and release them. Get them their shots and hope for the best. I am not a large scale operation.

Recently she picked up a few cats who were clearly feraland one that very likely wasnt. It had a collar with a phone number and was clearly well-cared for.

Recently I was called to an area overrun with orange cats. They were everywhere and I mostly ended up handing it over to professionals.

I did, however, manage to grab three very sweet cats. Two were terrified, skinny, a little beaten up, but overall friendly. The third one was a little gent.

He was tubby, well groomed, fish patterned collar, the works. My plan was to find his parents and drop him off with a warning to keep him in due to the large amount of traps being set. Then the little bastard sprayed the inside of my car to the freakin max.

He was also not neutered, and with the number of cats who resembled him in the vicinity, OP felt certain he was part of the problem.

So, apparently, he wasnt fixed. I couldnt really tell (longhair) and assumed he was so left my covers off. Big mistake!

We are accustomed to seeing embroidery made on a small scale, but artist Katerina Marchenko takes this art form and makes it lifesize. Using colorful strands of yarn, she renders massive human portraits on delicate tulle and showcases the endless possibilities this medium has to offer.

Marchenko shows how she approaches each piece via her social media. In these videos, we see her weave long and wide pieces of thread through the tulle using her hands (san needle). She begins by outlining the facial features like the eyes, nose, and mouth with black. Then, she adds various hues to create planes of color, filling the portrait with a sense of dimension. However, rather than trying to disguise her method, she highlights the thread medium by adding stylistic touches to these faces in the form of long loose threads which cascade down the face or radiate from the head.

These large-scale portraits possess similar qualities to expressive paintings. The way in which Marchenko manipulates the array of different colored threads mimics brushstrokes on a canvas. And by alternating between light and dark shades, she even models these human figures.

Fascinating Animation Compares the Size of Extinct Animas With Their Descendants

When we think of the size of extinct animals, our minds might go to the large T-Rex in Jurassic Park. As frightening as that looks, there are other creatures that, while long gone, would've been quite scary to encounter. YouTube channel WTD has created a digital animation showcasing the sizes of extinct animals alongside their extant descendants. Let's just say that some daunting creatures of today had much more intimidating relatives in the long-since past.

The video begins presenting the nuralagus, a yard-length relative of the rabbit. Things, however, quickly escalate from there. For example, while modern beavers are about 17 inches tall, their ancestor, the castoroides, was over three feet tall and eight feet long. The same goes for the Emperor penguin. At four feet tall, they are endearing, but the Anthropornis Nordenskjoeldi was six feet highso basically as tall as a professional basketball player.

Some threatening creatures currently in the world make an appearance alongside their ancestors. The 32-foot reticulated python is joined by the 45-foot titanoboa, while the saltwater crocodile, at about 10 feet long, is four times smaller than the Rhamphosuchus, which was 39 feet long.

The animation also features some creatures that, while almost similar in size to their descendants, had some very distinctive features. This includes the megaloceros' large antlers, which rival the size of a moose, or the arctotherium, a bear almost similar in height to the polar bear but with dark fur and native to Central and South America.

WTD, which is dedicated to collecting and making data comparison of all things in 3D, has also produced some other insightful animations comparing animals present and past. To stay up to date with its videos, you can subscribe to its YouTube channel.

My current gloves are cheap, rugged, fairly comfortable, but not attachment point. This is maybe my fourth or fifth pair? I usually will use them to death then replace. Tried a pair that was partly leather, but did not like it as much. The fingertip dexterity wasn't that good on that pair.

In March 2023, turmoil from the banking sector in the wake of Silicon Valley Banks collapse rocked the market with the types of issues that it hadnt experienced since the Great Recession. This was no fly-by-night operation; at the time, Silicon Valley Bank (SVB) was the sixteenth-largest bank in the United States and the bank of choice for many venture capitalists and startups.

This high-profile implosion and the lack of proper risk management in place at SVB led many individuals to question how safe their money was. Ultimately, the Federal Reserve stepped in to make the banks depositors whole, but the event still called the health of the banking system into question.

While not everyone is rushing to take their money out of banks, many more people are now at least aware of the risks of a single point of failure and are moving to diversify their assets by splitting them between multiple banks or even diversifying into alternatives like bitcoin and other cryptocurrencies for the first time.

Cult Conspiracy: US Charges Two in Plot Against Chinese Religious Group "IndyWatch Feed Spiritual"

Two Chinese nationals allegedly acting as agents for the Chinese government were apprehended in the United States following an undercover operation and were charged with trying to damage and topple an anti-communist Chinese religious group.

Authorities in Los Angeles arrested 70-year-old John Chen and 43-year-old Lin Feng on May 26th, where they were indicted for planning to revoke the tax-exempt status of the New York-based Falun Gong spiritual movement in an attempt to further undermine the exiled group.

The two Chinese nationals were both born in China but lived in Los Angeles at the time of their arrest. Chen is a US citizen, while Feng was described as a lawful permanent resident. Theyre facing charges of being unregistered agents of a foreign government, practicing money laundering, and bribing public officials.

Chen and Feng were caught after they initially urged the Internal Revenue Service (IRS) to revoke Falun Gongs non-profit tax status. Chen also described the organization as a gigantic mega cult in his whistleblower complaint that he filed to the tax agency last February, echoing the language used by the Chinese Communist Party (CCP) against the group.

After filing their complaint, an undercover police officer posed as an official for the IRS. According to prosecutors handling their case, Chen met with the officer at a restaurant north of New York City on May 14th.

One of two albums of theirs that clocks in at over 30 minutes, and you know what they say: if a Circle Jerks record lasts longer than a half hour, consult a physician immediately. Though its got some fun stuff going on at various points the Seven Dwarfs-esque whistling on the hook of the title track, and that magnificent country-fried guitar solo on Mrs. Jones come to mind it just isnt up to the snotty, rabid standard we know these fellas are capable of. Opinion: Quiet Confidence Has Nothing on Loud Insecurity "IndyWatch Feed Satire"

Spend enough time scrolling through Instagram or aimlessly clicking through Pinterest boards and youll see all kinds of advocacy for something called quiet confidence, where you recognize your strengths and feel comfortable in yourself without needing to prove to anyone else that you have value. Now thats all well and good for the wake up at 6, in bed by 10 crowd, but I tried quiet confidence and let me tell you, it pales compared to the rush of loud insecurity.

I was telling my therapist about how any criticism at work eats away at me to the point that Im running back and forth to the bathroom to soak paper towels in water to hide any signs of crying. She suggested I do some journaling to help me untangle my negative thought patterns. Now, I cant say this didnt work because I didnt bother trying it. Instead, I loudly mentioned every mistake made by my coworkers to help deflect any criticism. This has yielded far more satisfying results.

This isnt limited to the workplace. Ive also harnessed this power to turn all my interpersonal relationships into one-sided pleas for validation that inevitably implode when the other person has enough of me bringing up that I was wait-listed at Dartmouth any time Im around someone who I perceive to be smarter than me. Did I get in? Nope. But that doesnt mean I cant name-drop an Ivy League school to help puff up my ego for a minute!

I also have to shout out the internet and social media for making it so easy to broadcast my debilitating lack of self-esteem. After all, what better way to scrutinize my worth as a human than by giving me an endless number of platforms to express how I have no conception of myself as anything but a vessel for others scorn? And if I dont get enough likes on a post, Ill definitely follow it up with another one directly pleading for attention. How many is enough? I still dont know!

But heres what I do know: loud insecurity has helped me to realize my lifes purpose, which is to try and do all I can to be recognized as worthwhile and desirable without any sort of willingness to reflect on my flaws and work to change them through consistent effort. Thats pretty special, right?

In the control group, no intervention was applied. The Vancouver Scar Scale (VSS), Quick Disabilities of Arm, Shoulder and Hand (QuickDASH) functional score, and Visual Analog Scale (VAS) satisfaction scores were assessed preoperatively and postoperatively. Follow-up evaluations were scheduled at 1, 3 and 6 months after CTR surgery.RESULTS: The overall VSS score of the intervention group significantly improved at 3-6 months (-1.260, p = .040). Previous studies reported that ECa 233 enhanced memory retention and synaptic plasticity in the hippocampus of healthy rats. Because of this, we became curious whether ECa 233 has a therapeutic effect on the fear memory deficit in the triple transgenic Alzheimer's disease (3xTg-AD) model mice. Fear memory is a crucial emotional memory for survival that is found to be impaired in patients with early-onset Alzheimer's disease (AD). In this study, we orally administered ECa 233 (doses: 10, 30, and 100[Formula: see text]mg/kg) to 3xTg-AD mice, who were five months old, for 30 consecutive days. We found that ECa 233 prevented a cued fear memory deficit and enhanced hippocampal long-term potentiation (LTP) in 3xTg-AD mice. Subsequent proteomic and western blot analyses revealed increased expression levels of the molecules related to LTP induction and maintenance, including brain-derived neurotrophic factor (BDNF), tyrosine receptor kinase B (TrkB) and its network proteins, and extracellular signal-regulated kinase 1 and 2 (ERK1 and 2) in the hippocampi and amygdala of 3xTg-AD mice after ECa 233 pre-treatment. Our results indicate that ECa 233 is a promising potential herbal standardized extract that could be used in preventing the fear memory deficit and synaptic dysfunction before the early onset of AD.

Abstract:  BACKGROUND: Estrogen receptor-positive (ER) breast cancer is the most diagnosed subtype, with aromatase inhibitors (AIs) being one of the therapeutic drug types used in the clinic. However, endocrine resistance may develop after prolonged treatment, and different approaches, such as combining endocrine and targeted therapies, have been applied. Recently, we demonstrated that cannabidiol (CBD) induces anti-tumor actions in ERbreast cancer cells by targeting aromatase and ERs. Considering this, we studied, in vitro, whether CBD when combined with AIs could improve their effectiveness.METHODS: MCF-7aro cells were used and the effects on cell viability and on the modulation of specific targets were investigated.RESULTS: CBD when combined with anastrozole (Ana) and letrozole (Let) caused no beneficial effect in comparison to the isolated AIs. In contrast, when combined with the AI exemestane (Exe), CBD potentiated its pro-cell death effects, abolished its estrogen-like effect, impaired ERactivation, and prevented its oncogenic role on the androgen receptor (AR). Moreover, this combination inhibited ERKactivation, promoting apoptosis. The study of the hormonal microenvironment suggests that this combination should not be applied in early stages of ERbreast tumors.CONCLUSIONS: Contrary to Ana and Let, this study highlights the potential benefits of combining CBD with Exe to improve breast cancer treatment and opens up the possibility of new therapeutic approaches comprising the use of cannabinoids.

The Nervous System in the Classroom | Gary Sharpe "IndyWatch Feed Health"

From Gary Sharpe: The work of Dr Stephen Porges, and his Polvagal Theory, was instrumental in my own understanding and knowledge of the Nervous System, chronic illness and trauma, and from there, the human condition, and societal issues. It is what led me to glean that my own Idiopathic Parkinsons Disease diagnosis was actually me being stuck in the freeze or tonic immobility stress response.

Porges basic tenet is that our Nervous System is constantly evaluating the environment for cues of threats or safety, through a danger sense he calls neuroception. If our neuroception deems the situation to be safe, we can be in calm, relaxed, and socially engaged states. If it deems there is a threat, it activates fight or flight responses, or if the danger is perceived as sufficiently great, puts us into a state of freeze.

Note here the use of the term safe has nothing to do with health & safety or safe spaces it means feeling safe at a visceral, Nervous System level.

Porges has used his work as a lens to look at various aspects of modern life, such as the design of schools and hospitals, and found them wanting. Indeed, through this lens, many of our institutions seem almost designed to constantly trigger our sense of threat.

In his 2011 interview The Polyvagal Theory, Somatic Perspectives, Porges puts education systems and schools under the spotlight of his Nervous Systems work, explaining why modern schooling and educational methods are at odds with healthy Nervous System functioning. Here, we work through this important work.

However, even though we are discussing the classroom, for me the direct analogies carry over in a very straightforward way to any and all of our institutions where some folks have control or power over the lives of others. For example we could have talked about prisons, where we can straightforwardly swap out teacher for prison guard, and child for prisoner, or corporate employment, where we swap teacher for manager/boss, and child for employee, hospitals/healthcare (doctor and patients), government (elected politician and electorate), or caregiving (caregiver and cared for).

PMID: 37243404 Abstract Title:  Efficacy of cannabidiol in convulsive and nonconvulsive seizure types associated with treatment-resistant epilepsies in the expanded access program. Abstract:  Cannabidiol (CBD) Expanded Access Program (EAP), initiated in 2014, provided CBD (Epidiolex) to patients with treatment-resistant epilepsy (TRE). In the final pooled analysis of 892 patients treated through January 2019 (median exposure 694days), CBD treatment was associated with a 46%-66% reduction in median monthly total (convulsive plus nonconvulsive) seizure frequency. CBD was well tolerated, and adverse events were consistent with previous findings. We used pooled EAP data to investigate the effectiveness of add-on CBD therapy for individual convulsive seizure types (clonic, tonic, tonic-clonic, atonic, focal to bilateral tonic-clonic) and nonconvulsive seizure types (focal with and without impaired consciousness, absence [typical and atypical], myoclonic, myoclonic-absence) and epileptic spasms. CBD treatment was associated with a reduction in the frequency of convulsive seizure types (median percentage reduction, 47%-100%) and nonconvulsive seizure types and epileptic spasms (median percentage reduction, 50%-100%) across visit intervals through 144weeks of treatment. Approximately 50% of patients had50% reduction in convulsive and nonconvulsive seizure types and epileptic spasms at nearly all intervals. These results show a favorable effect of long-term CBD use in patients with TRE who may experience various convulsive and nonconvulsive seizure types. Future controlled trials are needed to confirm these findings.

Abstract:  Obesity is one of the principal public health concerns leading to disturbances in glucose and lipid metabolism, which is a risk factor for several chronic diseases, including insulin resistance, type 2 diabetes mellitus, and cardiovascular diseases. In recent years, it turned out that cannabidiol (CBD) is a potential therapeutic agent in the treatment of obesity and its complications. Therefore, in the present study, we used CBD therapy (intraperitoneal injections in a dose of 10 mg/kg of body mass for 14 days) in a rat model of obesity induced by a high-fat diet (HFD). Gas-liquid chromatography and Western blotting were applied in order to determine the intramuscular lipid content and total expression of selected proteins in the white and red gastrocnemius muscle, respectively. Based on fatty acid composition, we calculated de novo lipogenesis ratio (16:0/18:2n-6), desaturation ratio (18:1n-9/18:0), and elongation ratios (18:0/16:0, 20:0/18:0, 22:0/20:0 and 24:0/22:0), in the selected lipid fractions. Two-week CBD administration significantly reduced the intramuscular fatty acids (FAs) accumulation and inhibited de novo lipogenesis in different lipid pools (in the free fatty acid, diacylglycerol, and triacylglycerol fractions) in both muscle types, which coincided with a decrease in the expression of membrane fatty acid transporters (fatty acid translocase, membrane-associated fatty acid binding protein, and fatty acid transport proteins 1 and 4). Moreover, CBD application profoundly improved the elongation and desaturation ratios, which was in line with downregulated expression of enzymes from the family of elongases and desaturases regardless of the metabolism presented by the muscle type. To our knowledge, this study is the first that outlines the novel effects of CBD action on skeletal muscle with different types of metabolism (oxidative vs. glycolytic).

Abstract:  BACKGROUND: Cannabidiol (CBD) extracted from the cannabis plant is believed to have a medicinal value due to its neuroprotective effect via anti-inflammatory and antioxidant action. Recent behavioral studies in rats have reported that CBD mediates serotonin (5-HT1A) receptor action to improve motor dysfunction induced by dopamine (D2) receptor blockade. In particular, its effect on D2 receptor blockade in the striatum is an important function associated with neurological disorders resulting from various extrapyramidal motor dysfunctions. Dopaminergic neurodegeneration associated with this site is known for inducing Parkinson's disease (PD), which often affects the elderly. It is also known to cause drug-induced Parkinsonism. This study examines the ameliorating effect of CBD, which does not act directly on D2 receptors, against drug-induced motor dysfunction induced by the antipsychotic drug (haloperidol).METHODS: We created a drug-induced Parkinsonism model in zebrafish larvae using an antipsychotic drug (haloperidol). We evaluated the distance traveled and repetitive light-stimulation response. Furthermore, we examined whether administration of several concentrations of CBD ameliorates symptoms of the Parkinsonism model and compared its effects with those of antiparkinsonian drug ropinirole.RESULTS: CBD concentrations equal to half of haloperidol's resulted in an almost complete reversal of haloperidol-induced motor dysfunction, as measured by the distance traveled by the zebrafish and their response to light-stimulus. While ropinirole also significantly reversed haloperidol's effects at the same concentration as CBD, CBD was more effective than ropinirole.CONCLUSIONS: CBD-induced motor dysfunction improvement via D2 receptor blockade is a potential novel mechanism for the treatment of haloperidol-induced motor dysfunction.

Abstract:  INTRODUCTION: With the legalization of cannabis in multiple jurisdictions throughout the world, a larger proportion of the population consumes cannabis. Several studies have demonstrated anti-tumor effects of components present in cannabis in different models. Unfortunately, little is known about the potential anti-tumoral effects of cannabinoids in bladder cancer and how cannabinoids could potentially synergize with chemotherapeutic agents. Our study aims to identify whether a combination of cannabinoids, like cannabidiol and-tetrahydrocannabinol, with agents commonly used to treat bladder cancer, such as gemcitabine and cisplatin, can produce desirable synergistic effects. We also evaluated if co-treatment with different cannabinoids resulted in synergistic effects.METHODS: We generated concentration curves with several drugs, including several cannabinoids, to identify the range at which they could exert anti-tumor effects in bladder cancer cell lines. We tested the cytotoxic effects of gemcitabine (up to 100 nM), cisplatin (up to 100 M), and cannabinoids (up to 10 M) in T24 and TCCSUP cells. We also evaluated the activation of the apoptotic cascade and whether cannabinoids have the ability to reduce invasion in T24 cells.RESULTS: Cannabidiol,-tetrahydrocannabinol, cannabichromene, and cannabivarin reduce cell viability of bladder cancer cell lines, and their combination with gemcitabine or cisplatin may induce differential responses, from antagonistic to additive and synergistic effects, depending on the concentrations used. Cannabidiol and-tetrahydrocannabinol were also shown to induce apoptosis via caspase-3 cleavage and reduce invasion in a Matrigel assay. Cannabidiol and-tetrahydrocannabinol also display synergistic properties with other cannabinoids like cannabichromene or cannabivarin, although individual cannabinoids may be sufficient to reduce cell viability of bladder cancer cell lines.DISCUSSION: Our results indicate that cannabinoids can reduce human bladder transitional cell carcinoma cell viability, and that they can potentially exert synergistic effects when combined with other agents. Our in vitro results will form the basis for future studies in vivo and in clinical trials for the development of new therapies that could be beneficial for the treatment of bladder cancer in the future.

Abstract:  Synthetic cannabidiol (CBD) derivative VCE-004.8 is a peroxisome proliferator-activated receptor gamma (PPAR) and cannabinoid receptor type 2 (CB) dual agonist with hypoxia mimetic activity. The oral formulation of VCE-004.8, termed EHP-101, possesses anti-inflammatory properties and is currently in phase 2 clinical trials for relapsing forms of multiple sclerosis. The activation of PPARor CBreceptors exerts neuroprotective effects by dampening neuroinflammation in ischemic stroke models. However, the effect of a dual PPAR/CBagonist in ischemic stroke models is not known. Here, we demonstrate that treatment with VCE-004.8 confers neuroprotection in young mice subjected to cerebral ischemia. Male C57BL/6J mice, aged 3-4 months, were subjected to 30-min transient middle cerebral artery occlusion (MCAO). We evaluated the effect of intraperitoneal VCE-004.8 treatment (10 or 20 mg/kg) either at the onset of reperfusion or 4h or 6h after the reperfusion. Seventy-two hours after ischemia, animals were subjected to behavioral tests. Immediately after the tests, animals were perfused, and brains were collected for histology and PCR analysis. Treatment with VCE-004.8 either at the onset or 4h after reperfusion significantly reduced infarct volume and improved behavioral outcomes. A trend toward reduction in stroke injury was observed in animals receiving the drug starting 6h after recirculation. VCE-004.8 significantly reduced the expression of pro-inflammatory cytokines and chemokines involved in BBB breakdown. Mice receiving VCE-004.8 had significantly lower levels of extravasated IgG in the brain parenchyma, indicating protection against stroke-induced BBB disruption. Lower levels of active matrix metalloproteinase-9 were found in the brain of drug-treated animals. Our data show that VCE-004.8 is a promising drug candidate for treating ischemic brain injury. Since VCE-004.8 has been shown to be safe in the clinical setting, the possibility of repurposing its use as a delayed treatment option for ischemic stroke adds substantial translational value to our findings.

Abstract:  BACKGROUND: Prostate cancer is the second most frequently occurring carcinoma in males worldwide and one of the leading causes of death in men around the world. Recent studies estimate that over 1.4 million males are diagnosed with prostate cancer on an annual basis, with approximately 375,000 succumbing to the disease annually. With current treatments continuing to show severe side effects, there is a need for new treatments. In this study we looked at the effect of cannabis sativa extract, cannabidiol and cisplatin on prostate cancer cells, PC3.METHODS: In addressing the above questions, we employed the MTT assay to measure the antiproliferative effect on PC3 cells following treatment with varying concentrations of Cannabis sativa extract, cisplatin and cannabidiol. xCELLigence was also used to confirm the IC50 activity in which cells were grown in a 16 well plate coated with gold and monitor cell attachment. Caspase 3/7 activity was also measured using 96 well-plate following treatment. Western-blot and qRT-PCR was also used to measure the gene expression of tumour suppressor genes, p53, Bax and Bcl2. Animal studies were employed to measure the growth of PC3-mouse derived cancer to evaluate the effect of compounds in vivo.RESULTS: From the treatment with varying concentrations of Cannabis sativa extract, cannabidiol and cisplatin, we have observed that the three compounds induced antiproliferation of PC3 cancer cell lines through the activation of caspase 3/7 activity. We also observed induction of apoptosis in these cells following silencing of retinoblastoma binding protein 6 (RBBP6), with upregulation of p53 and bax mRNA expression, and a reduction in Bcl2 gene expression. The incidence is highest in adolescents, and the most common sites are the distal femur, proximal tibia and proximal humerus metaphyseal. Doxorubicin is the first-line chemotherapeutic agent for the treatment of osteosarcoma, but it has many side effects. Cannabidiol is a non-psychoactive plant cannabinoid cannabinol (CBD) that has been shown to be effective against osteosarcoma; however, the molecular targets and mechanisms of CBD action in osteosarcoma remain unclear.METHODS: Cell proliferation, migration, invasion and colony formation were analyzed using two drugs alone or in combination to evaluate their inhibitory effects on the malignant characteristics of OS cells. Apoptosis and the cell cycle were detected by flow cytometry. The synergistic inhibitory effect of doxorubicin/cannabidiol on tumors was also detected in nude mouse xenotransplantation models.RESULTS: Through analysis of two osteosarcoma cell lines, MG63 and U2R, it was found that the cannabidiol/doxorubicin combination treatment synergistically inhibited growth, migration and invasion and induced apoptosis, blocking G2 stagnation in OS cells. Further mechanistic exploration suggests that the PI3K-AKT-mTOR pathway and MAPK pathway play an important role in the synergistic inhibitory effect of the two drugs in osteosarcoma. Finally, in vivo experimental results showed that the cannabidiol/doxorubicin combination treatment significantly reduced the number of tumor xenografts compared to cannabidiol alone or doxorubicin alone.CONCLUSIONS: Our findings in this study suggest that cannabidiol and doxorubicin have a synergistic anticancer effect on OS cells, and their combined application may be a promising treatment strategy for OS.

However, CBD also has prominent anti-inflammatory and immunomodulatory effects; evidence exists that it could be beneficial in chronic inflammation, and even in acute inflammatory conditions, such as those due to SARS-CoV-2 infection. In this work, we review available evidence concerning CBD's effects on the modulation of innate immunity. Despite the lack so far of clinical studies, extensive preclinical evidence in different models, including mice, rats, guinea pigs, and even ex vivo experiments on cells from human healthy subjects, shows that CBD exerts a wide range of inhibitory effects by decreasing cytokine production and tissue infiltration, and acting on a variety of other inflammation-related functions in several innate immune cells. Clinical studies are now warranted to establish the therapeutic role of CBD in diseases with a strong inflammatory component, such as multiple sclerosis and other autoimmune diseases, cancer, asthma, and cardiovascular diseases.

After the isolation of CBD follows the discovery of CB1 and CB2 receptors and the main enzymatic components (diacylglycerol lipase (DAG lipase), monoacyl glycerol lipase (MAGL), fatty acid amino hydrolase (FAAH)). At the same time, the antioxidant potential of CBD is due not only to the molecular structure but also to the fact that this compound increases the expression of the main endogenous antioxidant systems, superoxide dismutase (SOD), and glutathione peroxidase (GPx), through the nuclear complex erythroid 2-related factor (Nrf2)/Keep1. Regarding the role in the control of inflammation, this function is exercised by inhibiting (nuclear factor kappa B) NF-B, and also the genes that encode the expression of molecules with a pro-inflammatory role (cytokines and metalloproteinases). The other effects of CBD on cognitive function and physical performance should not be excluded. In conclusion, the CBD market needs to be regulated more thoroughly, given the previously listed properties, with the mention that the safety profile is a very good one.

It has been shown that CBD can inhibit the proliferation of ovarian cancer cells, but the underlying specific mechanism is unclear. We previously presented the first evidence for the expression of leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1), a member of the immunosuppressive receptor family, in ovarian cancer cells. In the present study, we investigated the mechanism by which CBD inhibits the growth of SKOV3 and CAOV3 ovarian cancer cells, and we sought to understand the concurrent role of LAIR-1. In addition to inducing ovarian cancer cell cycle arrest and promoting cell apoptosis, CBD treatment significantly affected the expression of LAIR-1 and inhibited the PI3K/AKT/mTOR signaling axis and mitochondrial respiration in ovarian cancer cells. These changes were accompanied by an increase in ROS, loss of mitochondrial membrane potential, and suppression of mitochondrial respiration and aerobic glycolysis, thereby inducing abnormal or disturbed metabolism and reducing ATP production. A combined treatment with N-acetyl-l-cysteine and CBD indicated that a reduction in ROS production would restore PI3K/AKT/mTOR pathway signaling and ovarian cancer cell proliferation. We subsequently confirmed that the inhibitory effect of CBD on the PI3K/AKT/mTOR signal axis and mitochondrial bioenergy metabolism was attenuated by knockdown of LAIR-1. Our animal studies further support the in vivo anti-tumor activity of CBD and suggest its mechanism of action. In summary, the present findings confirm that CBD inhibits ovarian cancer cell growth by disrupting the LAIR-1-mediated interference with mitochondrial bioenergy metabolism and the PI3K/AKT/mTOR pathway. These results provide a new experimental basis for research into ovarian cancer treatment based on targeting LAIR-1 with CBD.

Abstract:  BACKGROUND: Although topical steroids constitute the first-line therapy for recurrent aphthous ulcers (RAUs), their long-term use often leads to candidiasis. Although cannabidiol (CBD) can be an alternative for pharmacologically managing RAUs due to its analgesic and anti-inflammatory in vivo effects, there is a lack of clinical and safety trials concerning its use. The aim of this study was to evaluate the clinical safety and efficacy of topical 0.1% CBD for managing RAU.METHODS: A CBD patch test was performed on 100 healthy subjects. CBD was applied on the normal oral mucosa of 50 healthy subjects 3 times/day for 7 days. Oral examination, vital signs, and blood tests were performed pre- and post-CBD use. Another 69 RAU subjects randomly received one of three topical interventions: 0.1% CBD, 0.1% triamcinolone acetonide (TA), or placebo. These were applied on the ulcers 3 times/day for 7 days. The ulcer and erythematous size were measured on day 0, 2, 5, and 7. Pain ratings were recorded daily. The subjects rated their satisfaction with the intervention and completed a quality-of-life questionnaire (OHIP-14).RESULTS: None of the subjects exhibited allergic reactions or side effects. Their vital signs and blood parameters were stable before and after the 7-day CBD intervention. CBD and TA significantly reduced ulcer size more than placebo at all time points. The erythematous size reduction was higher in the CBD intervention than the placebo on day 2, while TA reduced the erythematous size at all time points. The pain score in the CBD group was lower compared with placebo on day 5, whereas TA reduced pain more than placebo on day 4, 5, and 7. The subjects receiving CBD reported higher satisfaction than placebo. However, the OHIP-14 scores were comparable among the interventions.CONCLUSIONS: Topical 0.1% CBD reduced ulcer size and accelerated ulcer healing without side effects. CBD exerted anti-inflammatory effects in the early stage and an analgesic effect in the late RAU stage. Thus, topical 0.1% CBD might be more appropriate for RAU patients who decline to take topical steroids, except for cases where CBD is contraindicated.TRIAL REGISTRATION: Thai Clinical Trials Registry (TCTR) Number TCTR20220802004. However, several authors have reported contradictory findings concerning the magnitude and direction of CBD-mediated effects. For example, CBD treatment can increase, decrease, or have no significant effect on autophagy and apoptosis. These variable results can be attributed to the differences in the biological models, cell types, and CBD concentration used in these studies. This review focuses on the mechanism of regulation of autophagy and apoptosis in inflammatory response and cancer by CBD. Further, we broadly elaborated on the prospects of using CBD as an anti-inflammatory agent and in cancer therapy in the future.

Abstract:  Lung cancer remains the most chronic form of cancer and the leading cause of cancer mortality in the world. Despite significant improvements in the treatment of lung cancer, the current therapeutic interventions are only partially effective, necessitating the continued search for better, novel alternative treatments. Angiogenesis and cancer stem cells play a central role in the initiation and propagation of cancers. Tumor angiogenesis is triggered by an angiogenic switch when pro-angiogenic factors exceed anti-angiogenic components. Although many anti-angiogenic agents are used in cancer treatment, there are therapeutic limitations with significant side effects. In recent years, cannabinoids have been investigated extensively for their potential anti-neoplastic effects. Our previous findings showed that cannabidiol (CBD) could impede tumor growth in mouse models of melanoma and glioblastoma. Importantly, CBD has been suggested to possess anti-angiogenic activity. In this study, we tested, for the first time, inhalant CBD in the treatment of heterotopic lung cancer and whether such potential effects could reduce cancer stem cell numbers and inhibit tumor angiogenesis. We implanted NCI H1437 human lung cancer cells in nude mice and treated the mice with inhalant CBD or placebo. The outcomes were measured by tumor size and imaging, as well as by immunohistochemistry and flow cytometric analysis for CD44, VEGF, and P-selectin. Our findings showed that CBD decreased tumor growth rate and suppressed expression of CD44 and the angiogenic factors VEGF and P-selectin. These results suggest, for the first time, that inhalant CBD can impede lung cancer growth by suppressing CD44 and angiogenesis.

How Excess Iron Raises Your Risk for Alzheimer's "IndyWatch Feed Health"

While dietary iron is essential for optimal health1 being a key part of proteins and enzymes and playing an important role in energy production and the regulation of cell growth and differentiation, among other things too much iron in your body can have serious ramifications.2 One of the most important roles of iron is to provide hemoglobin (the protein in red blood cells) a mechanism through which it can bind to oxygen and carry it throughout your tissues.

Without proper oxygenation, your cells cannot function properly and eventually die. Common symptoms of insufficient iron include fatigue, decreased immunity or iron deficiency anemia, which can be serious if left untreated.

However, your body has a very limited capacity to excrete iron, which means it can build up in your tissues and organs. This is problematic, as iron is a potent oxidizer, capable of damaging tissues, including your vascular system and brain, thereby raising your risk for both heart disease and dementia.

Excess Iron 'Rusts' Your Brain

You're probably familiar with the fact that Alzheimer's disease is associated with a buildup of amyloid beta plaque in the brain. According to recent research3,4 from the Netherlands, buildup of iron, causing a rusting effect in the brain, also plays an important role and is common in most Alzheimer's patients. As noted by the authors:

"In the presence of the pathological hallmarks of [Alzheimer's disease], iron is accumulated within and around the amyloid-beta plaques and neurofibrillary tangles, mostly as ferrihydrite inside ferritin, hemosiderin and magnetite.

The co-localization of iron with amyloid-beta has been proposed to constitute a major source of toxicity. Indeed, in vitro, amyloid-beta has been shown to convert ferric iron to ferrous iron, which can act as a catalyst for the Fenton reaction to generate toxic free radicals, which in turn result in oxidative stress."

Addressing excess iron may therefore be an effective treatment option. I graduated from the University of Toronto School of Architecture [in 1978] and then, shortly after, moved to Santa Fe. I was living in a home that was far from ideal. It was new.

Abstract:  Cannabidiol (CBD) (25 mg/kg peroral) treatment was shown to improve metabolic outcomes in ovariectomized (OVX) mice deficient in 17-estradiol (E2). Herein, CBD effects on intestinal and hepatic bile acids (BAs) and inflammation were investigated. Following RNA sequencing of colon tissues from vehicle (VEH)- or CBD-treated sham surgery (SS) or OVX mice (= 4 per group), differentially expressed genes (DEGs) were sorted in ShinyGO. Inflammatory response and bile secretion pathways were further analyzed. Colon content and hepatic BAs were quantified by LC-MS (= 8-10 samples/group). Gut organoids were treated with CBD (100, 250, 500M) with or without TNFand lipopolysaccharide (LPS) followed by mRNA extraction and qPCR to assess CBD-induced changes to inflammatory markers. The expression of 78 out of 114 inflammatory response pathway genes were reduced in CBD-treated OVX mice relative to vehicle (VEH)-treated OVX mice. In contrast, 63 of 111 inflammatory response pathway genes were increased in CBD-treated sham surgery (SS) mice compared to VEH-treated SS group and 71 of 121 genes were increased due to ovariectomy. CBD did not alter BA profiles in colon content or liver. CBD repressedandexpression in intestinal organoids in a dose-dependent manner. In conclusion, CBD suppressed colonic inflammatory gene expression in E2-deficient mice but was pro-inflammatory in E2-sufficient mice suggesting CBD activity in the intestine is E2-dependent.

Since recent reports have suggested that cannabidiol (CBD) may assist in the treatment of inflammatory diseases, our objective was to examine the effect of CBD on cytokine storm-induced cardiac and renal injury using the lipopolysaccharide (LPS)-induced sepsis mouse model.At 8 weeks of age, mice were randomly assigned to receive CBD (15mg/kg) or vehicle one hour before a single injection of either phosphate-buffered saline or LPS (10mg/kg) for an additional 24h.Our results show that CBD improves cardiac function and reduces renal injury in a mouse model of cytokine storm. Moreover, our data indicate that CBD significantly reduces systemic and renal inflammation to contribute to the improvements observed in a cytokine storm-model of cardiac and renal injury.Overall, the findings of this study suggest that CBD could be repurposed to reduce morbidity in patients with cytokine storm particularly in severe infections such as sepsis.
